基于网络药理学的熟地黄环烯醚萜类成分治疗类风湿性关节炎作用机制的初步探究

摘    要:目的:对熟地黄环烯醚萜类成分治疗类风湿性关节炎(RA)的多成分、多靶点、多途径的作用机制进行初步探究。方法:通过文献查找和TCMSP数据库筛选出候选成分,利用PubChem数据库、TTD数据库和DrugBank数据库筛选出各成分所作用的并与RA相关的靶点,再将靶点信息导入David数据库,得到重要靶点和通路信息。最后使用Cytoscape3.7.0软件对筛选后的成分、靶点和通路建立网络图。结果:由网络图的分析报告得知肿瘤坏死因子(TNF)、白细胞介素(IL)-1β、IL-6和核因子-κB(NF-κB)抑制蛋白-α(IκBα)是熟地黄环烯醚萜类成分作用的关键靶点;NF-κB信号通路、Toll样受体信号通路、TNF信号通路、细胞凋亡是其产生抗RA活性的主要通路;说明熟地黄环烯醚萜类成分的“多成分-多靶点-多通路”的作用机制。结论:熟地黄环烯醚萜成分主要是通过细胞因子和细胞调节发挥抗RA活性。

Abstract:Objective: To explore the mechanism of multi-component, multi-target and multi-pathway of iridoids from Rehmanniae Radix Praeparata in the treatment of rheumatoid arthritis(RA). Methods: Candidate ingredients were screened by literature search and TCMSP database. The RA-related targets prediction and filtering for each candidate ingredient were conducted by PubChem database, TTD database and DrugBank database. Then target information was imported into David database to obtain important target and pathway information. Finally, the network of the candidate ingredients, targets and pathways was constructed by Cytoscape 3.7.0 software. Results: Analysis report of network diagram showed that TNF, IL-1β, IL-6, and IκBα were the important targets for the effect of iridoids from Rehmanniae Radix Praeparata. NF-κB signaling pathway, Toll-like receptor signaling pathway, TNF signaling pathway and apoptosis are the main pathways in the treatment of RA. The mechanism of "multi-component-multi-target-multi-pathway"of iridoids from Rehmanniae Radix Praeparata was illuminated. Conclusion: The iridoids component of Rehmanniae Radix Praeparata mainly exerts its anti-RA activity through cytokines and cellular regulatory pathways, which lays a foundation for the subsequent experiment to verify its mechanism of action.
