摘    要:肿瘤细胞耐药性是临床实践中肿瘤治疗失败的主要原因。近年来的研究认为,肿瘤细胞的耐药性可能是由于非遗传适应性机制如细胞可塑性所导致,而非基因突变引起。细胞可塑性特性致使肿瘤细胞在药物耐受与药物敏感的表型或状态之间可逆切换,进而逃逸药物攻击并存活下来。因而,细胞可塑性及其机制研究可为逆转肿瘤细胞耐药性提供新靶点、新策略。为此,本文简要介绍肿瘤细胞耐药性的非遗传适应性机制、药物诱导细胞可塑性现象的具体表现、药物耐受机制与靶向治疗策略。

Abstract:The therapeutic resistance is the main cause of failure in clinical practice of cancer treatment.Recent studies suggest that therapeutic resistance in cancer may be due to non-genetic adaptive mechanisms such as cell plasticity rather than genetic mutations.The properties of cellular plasticity enable tumor cells to reversibly switch their phenotypes or states between drug tolerance and drug-sensitive,and then escape and survive the therapeutic challenge.Therefore,the researches of cell plasticity and the corresponding mechanism could provide new targets and new strategies for reversing therapeutic resistance in cancer.To this end,this article briefly explores the non-genetic adaptive mechanism of therapeutic resistance,as well as the features,tolerance mechanism and therapeutic revelations for drug-induced cell plasticity.
