五味子丙素体外抗胃癌细胞系AGS的活性

目的：探讨中药五味子中五味子丙素的体外抗胃癌活性。方法：在胃癌细胞AGS中，采用MTT法和集落形成实验测定五味子丙素对细胞的生长抑制活性，采用流式细胞仪测定其诱导细胞凋亡的活性，采用Western blot法测定其对凋亡蛋白表达的影响。并用MTT法测定其对不良反应较大的传统胃癌一线化疗药物5-FU的增敏活性。结果：五味子丙素对胃癌细胞AGS生长抑制的IC50为（35.73±0.28）μmol/L，并能浓度剂量依赖性地抑制细胞集落形成，诱导细胞凋亡，上调凋亡蛋白Cleaved-PARP，下调抗凋亡蛋白Bcl-2的表达（P＜0.05）。结论：五味子丙素对胃癌细胞具有较好的抑制生长和诱导凋亡的活性。

Anti-gastric cancer activity of schisandrin C in AGS

Objective: To investigate in vitro anti-gastric cancer activity of schisandrin C. Methods: MTT assay and colony formation assay were used to examine the growth inhibitory activity of schisandrin C in AGS cells. After treatment with different concentrations of schisandrin C, the apoptosis of AGS cells was detected by flow cytometer. Meanwhile, Western blot was performed to detect the expression of apoptotic proteins. 5-FU, a traditional first-line chemotherapy drug for gastric cancer, MTT assay was used to detect the synergism with 5-FU. Results: The half maximal inhibitory concentration (IC50) of schisandrin C against AGS cells was (35.73± 0.28) μmol/L, which could inhibit cell colony formation, induce cell apoptosis, up-regulate the apoptotic protein Cleaved-PARP, and down-regulate the expression of anti-apoptotic protein Bcl-2 in a dose-dependent manner. Conclusion: Schisandrin C has stronginhibitory activity and induces apoptosis on gastric cancer cells.
