钩端螺旋体DNA限制性内切酶图谱分析

摘    要:七日热和波摩那各型标准株及一些野生菌株用EcoRI等六种限制性内切酶消化分析。结果表明,不同型菌株具有完全不同的酶切图谱;来自不同地域、不同时期、不同宿主的同型野生株酶谱一致,与它们相应的标准菌株没有差别或仅细小的差别。说明钩端螺旋体具有血清型特征性限制性内切酶图谱,而且是相当稳定的。我们认为限制性内切酶图谱分析是一种比较灵敏、特异、快速、简便的钩端螺旋体分类鉴定方法,还可用于分子流行病学调查。

Abstract:The genomes of reference strains belonging to serogroups Hebdomadis and Pomona and fieldisolates belonging to serovars hebdomadis and pomona were analyzed by six restriction endonucleases '. The results showed obvious differences among reference strains. Field isolates belonging to same serogroup from various areas, periods and hosts could not differ from each other. And there was a tiny difference in high molecular regions between field isolates serovar hebdomadis and their reference,strain. However,field isolates of serovar pomona and their reference strain could not be distinguished by restriction endonuclease analysis (REA) with all six endonucleases. That showed that REA is a sensitive, specific method for classification of Leptospira. It may be applyed to molecular epidemiologic investigation of Leptospirosis.
