石棉微粒大鼠肺纤维化模型的制作及其意义

摘    要:目的:通过本实验为肺纤维化的发病机理、支端病理、药物筛选的研究提供一种简便的动物模型。方法:采用<5μm直径的石棉微粒直接给大鼠灌肺,分别在2,4,6,8周处死大鼠,用HE及改良的Gommor结缔组织特殊染色,在光镜下观察。结果:可见到大量的炎细胞浸润,吞噬细胞活跃,同时有相当数量的吞噬细胞落入肺泡腔内,肺泡壁有很多绿色的纤维组织增生。结论:本方法简便、成功率高、病理改变典型,对其发病机理、支端病理、药物筛选的研究提供了有利的实验条件。

Abstract:Objective:To study the machenism of the pathological change and pathology of fibrosis and to observe the effect of the drug,the model of pulmonary fribrosis of mouse was made.Methods:The asbestos particles (<5 mm in diameter) were poured into the lungs of mice.Then the mice were killed after two,four, six and eight weeks seperately.The specimens were dyed with the special method of HE and improved Gommor and the connective tissue and the samples were observed by photoscope.Results:A lot of inflammatory cells diffusing into the lungs were found.The function of phagocyte was obviously enhanced.And the large numbers of phagocytes intruded themselves into the alveoli.There was a lot of green fibrous tissues around the small blood vessels and in the walls of bronchiole and alveoli.Conclusion:The results suggested that the model was useful in the sutdy of the pulmonary fibrosis because the pathological changes were typical and the method was simple,stable and effective.
