孕期摄食限制所致IUGR子代大鼠高脂饮食下肾脏结构与功能改变

摘    要:目的:探讨高脂饮食对孕期摄食限制所致宫内发育迟缓(IUGR)子代大鼠肾脏结构功能的影响。方法:Wistar孕鼠随机分为对照组(CN)和摄食限制(FR)组,自孕11d至足月产分别给予自由进食和摄食限制(每天食物摄入量为正常对照组的50%)。仔鼠断奶后给予高脂饮食至20周,麻醉处死动物,检测子代大鼠肾重、肾脏指数(肾重/体重);观察肾脏组织病理学变化;取血清,检测血肌酐(SCr)、尿素氮(BUN)、肾素活性(PRA)、血管紧张素Ⅱ(AngⅡ)水平;RT-PCR检测肾脏组织肾素-血管紧张素系统(RAS)组分的mRNA表达。结果:高脂饮食下,FR组雌性及雄性子代大鼠SCr含量较正常对照组均显著升高(P<0.01),肾脏出现肾小球增生、硬化、间质炎性细胞浸润等病理组织学改变,血浆AngⅡ均显著下降(P<0.01);雌性PRA下降(P<0.01),雄性无变化;肾脏RAS mRNA表达不一,且存在性别差异。结论:高脂饮食下,孕期摄食限制所致IUGR子代大鼠成年后出现肾损伤表型,高脂饮食下存在IUGR子代肾脏损伤易感的现象。

Abstract:Objective:To investigate high-fat diet effects on renal morphology and structure of IUGR offspring rats induced by prenatal food restriction.Methods:Maternal rats were randomly divided into control and diet restriction group,which were treated with food ad libitum or diet restriction from gestational day 11 to full-term delivery respectively.All pups were weaned to an ad libitum high-fat diet.At the 20 weeks age,the pups were sacrificed,kidney weight and index,the morphology of offspring kidney,serum creatinine(SCr),blood urea nitrogen(BUN),angiotensinⅡ(AngⅡ)levels and mRNA of renin-agiotensin system(RAS)components in kidney were observed.Results:Under high-fat diet,SCr level increased significantly(P<0.01)and Serum AngⅡ decreased obviously(P<0.01)in both male and female offsprings in FR group compared with control,accompanied with morphological insults such as glomerular hypertrophy,sclerosis and interstitial inflammatory infiltration.RAS mRNA expressions changed with gender differences.Conclusion:High-fat diet results in the renal dysfunction and morphological damage in adult IUGR offspring induced by prenatal food restriction,these results demonstrate the susceptibility of kidney injury of adult IUGR offspring rats on a high-fat diet.
