摘    要:目的探讨在不同环境中观察膀胱癌细胞表达癌干细胞相关标志物CD44、CD133的变化情况,为进一步研究人体膀胱癌干细胞提供背景资料和前期数据。方法采用免疫组织化学与流式细胞术等技术,以膀胱癌5637细胞系为研究对象,通过建立体外培养、皮下移植瘤以及裸鼠淋巴结转移模型等模拟不同的生长微环境,观察CD44、CD133等癌干细胞相关标志物的表达情况。结果在体外培养环境中,膀胱癌5637的细胞膜蛋白CD44阳性表达,CD133则阴性表达;膀胱癌裸鼠移植瘤中CD44阳性表达,而CD133弱阳性表达。与体外培养体系比较,流式细胞术检测发现膀胱癌细胞5637中的CD44阳性表达水平无显著变化,而CD133阳性表达水平显著高于体外培养体系(P〈0.01);在裸鼠淋巴结转移模型中,转移淋巴结均有CD44、CD133阳性表达。结论体内外环境的改变可以调控膀胱癌细胞中部分癌干细胞相关标志物的变化,也提示膀胱癌5637细胞系存在癌干细胞。

Abstract:Objective To investigate the expression of cancer stem cell makers CD44 and CD133 in bladder cancer in different environments to provide background information and preliminary data for the further study of human bladder cancer.Methods Cell used bladder cancer 5637 cell line,the different environment(niche)included cell culture system,the model of human bladder cancer xenograft,the metastatic lymph node model of bladder cancer,CD44 and CD133 were detected by immunohistochemical staining and flow cytometry respectively.Results The bladder cancer 5637 cell line in vitro,CD44 were strong positive,CD133 was negative.In the model of xenograft,CD44 was positive,CD133 was low positive.Compared with the bladder cancer 5637 cell line in vitro,the positive expression of CD44 was no change and the positive expression of CD133 was much higher in the model of human bladder cancer xenograft.In the metastatic lymph node model,CD44,CD133 were positive.Conclusion The expression of bladder cancer stem cell markers are regulated by the different environments.Bladder cancer stem cells exist in the 5673 cell line.
