Abstract:Objective To explore and improve the skill points of heterotopic small bowel transplantation(SBT) models in mice,and construct the stable model for small bowel transplantation research.Methods 53 segmental small bowels of Balb/C mice were transplanted into abdominal cavities of C57BL/6 mice.Several skill points were improved: to abridge length of the transplanted small bowel and portal vein pedicle;to make elliptical incision on front wall of the donor ventral aorta with suture needle puncture way and form the anterior wall incision of inferior caval vein with syringe needle puncture way;to anastomose the vessels with continuous sutures;to reconstruct the intestinal tract through ligating proximal end of graft intestine and end-to-side anastomosis of distal end of graft intestine with recipient jejunum.Results 46 mice survived 7days in 53 heterotopic SBT cases.The successful rate was 86.8%.In experiments harvesting time of donors was 45±10 mins.and the graft cool ischemic time was 25±5 mins,recipient operation time was 56±7 mins,in which the abdominal aorta and IVC blockage time was 33±8 mins.The operations failed in 7cases,due to massive hemorrhage(1 case),long anastomose time and crural paralysis(3 cases),intestinal obstruction(1 case),anastomosis stenosis(1case),and anesthetic accident(1 case).Conclusions Improvement of the skill points is facilitated establishment of stable SBT model for science research.
