摘    要:目的:建立测定石蒜中加兰他敏含量的HPLC方法。方法:采用ODS色谱柱(46mm×150mm,5μm),以磷酸盐缓冲液(pH3~4)甲醇(93∶7)为流动相,流速10mL·min-1,二极管阵列检测器于289nm测定。结果:加兰他敏在3~30μg·mL-1线性关系良好(r=0.9997),最低检测限为0.30ng,测定加兰他敏的平均回收率为99.5%,RSD0.5%。结论:方法操作简便,结果可靠,重复性好,专属性强,将为石蒜的质量控制提供科学依据。

Abstract:OBJECTIVE: To establish a method for determination of galanthamine in Lycoris radiata. METHOD: HPLC separation was carried on a column of ODS (4.6 mm x 150 mm, 5 microm), with the mobile phase of phosphate buffer (pH = 3-4)-methanol (93:7) at the flow rate of 1.0 mL x min(-1), and the detection wavelength was set at 289 nm. RESULT: Galanthamine revealed linearity within the range of 3-30 microg x mL(-1) (r = 0.9997), the detection limit was 0.3 ng. The average recovery was 99.5% (RSD = 0.5%). CONCLUSION: The method is easy to operate and the results of the determination are accurate, it can be used to evaluate the quality of L. radiata.
