人胚胎甲状腺褪黑素受体mRNA的检测

摘    要:目的:检测胚胎甲状腺中是否表达褪黑素受体。方法:取人胚胎甲状腺,抽提总RNA并用RT-PCR方法检测其mRNA,阳性产物用自动测序仪测序。结果:人胚胎甲状腺总RNA以MT1及MT2两种引物进行的RT-PCR产物电泳呈现阳性条带,测序结果显示扩增产物与人类褪黑素受体cDNA相吻合。结论:人胚胎甲状腺组织中存在有褪黑素MT1,MT2两种受体的mRNA,表明该组织中有褪黑素MT1,MT2受体的表达。

Abstract:Objective: To make it clear whether the thyroid in human embryo express melatonin(MEL) receptor or not. Methods: Thyroids was collected to obtain total RNA and the mRNA of melatonin MT1 or MT2 was detected by RT PCR method. The positive production was sequenced with auto sequencing instrument. Results: The electrophoretic results of RT PCR product by MT1 and MT2 primer were all positive. Sequencing results indicated that the positive product coincided with cDNA of human melatonin receptor. Conclusion: There exists melatonin receptor mRNA in human embryo thyroid, either MT1 and MT2, which shows human embryo thyroid express both MT1 or MT2 receptor.
