平肝降压丸改善肾性高血压大鼠胰岛素抵抗的实验研究

摘    要:目的研究平肝降压丸对肾性高血压大鼠胰岛素抵抗(IR)的影响。方法将50只Wistar将50只Wistar大鼠随机分为空白对照组、模型对照组、卡托普利组和平肝降压丸低、高剂量组各10只。除空白对照组外,其余4组均采用缩窄肾动脉造成肾性高血压大鼠,同时给予高糖高脂饲料喂养4周,形成肾性高血压IR动物模型,空白对照组大鼠仅游离肾动脉,不予缩窄,同时给予常规饲料喂养。从第5周开始给药,平肝降压丸低、高剂量组(分别为1.8g/kg、3.6g/kg)及卡托普利组(5mg/kg),每日灌胃1次,容量为1mL/100g体质量,空白对照组及模型对照组给予等容积纯净水,连续4周。观察给药前后各组大鼠血压、血糖、空腹胰岛素(FINS)、胰岛素敏感指数(ISI)、血清甘油三酯(TG)和血清总胆固醇(TC)的变化。结果与空白对照组给药前比较,模型对照组,平肝降压丸低、高剂量组及卡托普利组血压均升高(P0.01)。给药后2、4周平肝降压丸低、高剂量组和卡托普利组血压均较模型对照组降低(P0.05,P0.01)。与空白对照组给药前比较,模型对照组,平肝降压丸低、高剂量组及卡托普利组血糖均升高(P0.01)。给药后2周,平肝降压丸高剂量组和卡托普利组血糖较模型对照组降低(P0.05)。给药后4周,平肝降压丸低、高剂量组和卡托普利组血糖较模型对照组降低(P0.01)。与模型对照组比较,平肝降压丸低、高剂量组和卡托普利组FINS均降低(P0.01),平肝降压丸低、高剂量组和卡托普利组ISI均升高(P0.01)。与模型对照组比较,平肝降压丸低、高剂量组和卡托普利组TC、TG含量均降低(P0.05,P0.01)。结论平肝降压丸在降低肾性高血压大鼠血压的同时,提高ISI,改善IR,为临床防治高血压病IR提供了必要的药效学依据。

Experimental study of Pinggan Jiangya pill on insulin resistance in renal hypertensive rats

Abstract:Objective To observe the effect of Pinggan Jiangya pill on insulin resistance(IR)in renal hypertensive rats.Methods 50 rats were randomly divided into control group,model group,positive drug Captopril group,low dose Pinggan Jiangya pill group and high dose Pinggan Jiangya pill group.Renal hypertensive rat model were established by use of narrowing renal artery,providing high glucose diet feeding for 4 weeks at the same time to form a hypertensive rat model with insulin resistance.Different administration in each group were continuously processed for 4 weeks,then the variation of blood press(BP)、fasting blood glucose(FBG),fasting serum insulin(FINS),insulin sensitivity index(ISI),serum triglyceride(TG)and total cholesterol(TC)and blood lipid were determined before and after administration.Results Compared with control group,the level of blood pressure in model group,positive drug Captopril group,low dose Pinggan Jiangya pill group and high dose Pinggan Jiangya pill group was increased(P0.01).The level of blood pressure in positive drug Captopril group,low dose Pinggan Jiangya pill group and high dose Pinggan Jiangya pill group was reduced for 2 and 4 weeks of treatment compared with model group(P0.05,P0.01).Compared with control group,the level of FBG in model group,positive drug Captopril group,low dose Pinggan Jiangya pill group and high dose Pinggan Jiangya pill group was increased(P0.01).The level of FBG in positive drug Captopril group and high dose Pinggan Jiangya pill group was reduced for 2 weeks of treatment compared with model group(P0.05).The level of FBG in positive drug Captopril group,low dose Pinggan Jiangya pill group and high dose Pinggan Jiangya pill group was reduced for 4 weeks of treatment compared with model group(P0.01).FINS in positive drug Captopril group,low dose Pinggan Jiangya pill group and high dose Pinggan Jiangya pill group was reduced compared with model group(P0.01).ISI in three groups was increased(P0.01).Compared with model group TC and TG in positive drug Captopril group,low dose Pinggan Jiangya pill group and high dose Pinggan Jiangya pill group was reduced(P0.01,P0.05).Conclusion Pinggan Jiangya pill can reduce the level of blood pressure of renal hypertensive rats with insulin resistance at the same time,raise insulin sensitivity index,improve insulin resistance,and provide the evidence of pharmacology for clinical prevention and treatment on Insulin resistance.
