高血压患者血管紧张素转换酶基因多态性分析

摘    要:目的探讨血管紧张素转换酶基因多态性与北京房山地区高血压遗传易感相关性。方法应用聚合酶链反应(PCR)技术,检测287例高血压患者和316例正常血压对照者的ACE基因多态性。结果群体相关分析结果表明,高血压组(包括原发性高血压和合并空腹血糖升高的高血压患者)ACE基因的D等位基因及其纯合基因型(D/D)频率较正常血压对照组明显升高(x2=4.14,P<0.05)。原发性高血压患者组ACE基因缺失纯合基因型频率较高血压伴有空腹血糖升高组显著增加,其频率分别为30.8%和19.2%(x2=4.45,P<0.05),而高血压伴空腹血糖升高者与对照者间D/D纯合基因型频率无显著差异。结论ACE基因插入/缺失多态性可能与房山地区汉族原发性高血压相关联。

Abstract:Objective To investigate whether the polymorphism in the angiotensin convertingenzyme gene is associated with hypertension. Methods A case-control study was conduced. Thedeletion/insertion polymorphism of the ACE gene was detennined by polymerase chain reaction (PCR)in the Chinese population including 287 hypeFtensives and 316 normotensive controls. Results Thefrequencies of D allele and DD genotype distribution were significantly increased in hypertensivepopulation (including essential hypertension (EH) and hypertensive with elevated fastin plasma glucose,(HT2) as compared with normotensive controls (x2=4. 14,P<0.05), The difference of DD genotypedistribution of the ACE gene between EH and NT were also statistically significant (x2=8.22, P<0.05);The frequencies of DD genoboe of the ACE gene in EH was higher than that in HT3 (30.8% vs 19.2%,x2=4.45, P<0.05). But no difference of DD genotype was found between HT2 and NT (x2=0.059,P=0.9). Conclusion The results suggested that D/I polymorphism of the ACE gene might be associatedwith hypertension in the Han population of Fangshan area.
