胶体金免疫层析法在鼠疫监测应用

摘    要:目的了解胶体金免疫层析法(GICA)快速诊断鼠疫试剂的应用效果。方法在鼠疫监测中用间接血凝(IHA)微量法和胶体金免疫层析法检测鼠型动物血清中鼠疫F1抗体;选IHA微量法初筛时发生凝集的鼠型动物血清,用GICA检测鼠疫F1抗体,设立IHA微量法抑制试验对照;在溶血血清中加入鼠疫阳性参考血清,用GICA检测鼠疫F1抗体,设立IHA对照;用鼠疫菌EV76株免疫鼠类建立动物模型,用GICA检测鼠疫F1抗体和F1抗原,分别设立IHA和反向间接血凝(RIHA)对照。结果 IHA微量法和GICA检测鼠型动物血清237份,两种方法检测结果一致,全部阴性;GICA及IHA微量法抑制试验检测IHA微量法初筛时发生凝集的鼠型动物血清57份,两种方法结果一致,均为阴性,证实初筛结果为非特异性凝聚;GICA检测含鼠疫鼠疫阳性参考血清的鼠型动物溶血血清45份,结果全部阳性,而IHA对照结果无法判断;用GICA检测动物模型中鼠类血清25份和鼠类脏器18份,结果分别与IHA和RIHA对照一致。结论鼠疫胶体金试剂使用简便、快速、特异、敏感、便于鼠疫监测现场和鼠疫应急应用。

Results of application of gold-immunochromatography assay in monitoring of plague and establishment of animal model

Abstract:Aim To understand the results of application of gold-immunochromatography assay(GICA)dipstick for rapid detection of Yersinia pestis.Methods F1 antibodies to Yersinia pestis were detected by IHA and GICA;controls were set up by inhibited test of IHA ;F1 antibodies of hemolytic serum mixed with serum positive for plague were detected by GICA,IHA control was also set up ;rats were immunized by EV76 strain of Yersinia pestis and animal model of plague was set up,F1 antibodies and F1 antigens were detected by GICA,IHA and RIHA controls were set up respectively.Results There 237 serum samples detected both by IHA and GICA were all negative;57 serum samples agglutinated during primary screening by IHA were also negative after detected by IHA and GICA,suggesting non-specific agglutination;45 hemolytic sera containing sera positive for Yersinia pestis detected by GICA were all positive.Results of detection of 25 sera and 18 viscera of animal model detected by GICA were in accordance with controls detected IHA and RIHA.Conclusion The diagnosis of plague using gold-immunochromatography assay issimple,rapid,specific and sensitive and suitable for monitoring pague in the field.
