| 摘 要： | 目的制备人心肌型脂肪酸结合蛋白(Heart type fatty acid-binding protein;H-FABP)单克隆抗体。方法(1)应用淋巴细胞杂交瘤技术,将重组人H-FABP腹腔注射免疫BALB/C小鼠。(2)将鼠脾细胞与鼠骨髓瘤细胞(SP2/0)融合、培养。通过HAT和间接ELISA法筛选、鉴定制备稳定分泌抗人H-FABP单克隆抗体的杂交瘤细胞株。(3)利用Protein G Sepharose 4 Fast Flow亲合纯化体内诱生法制备的腹水抗体,并对抗体特性进行鉴定。结果得到7株特异性识别人H-FABP单克隆抗体,并纯化两株亲合力高、空间位点远的单克隆抗体。结论成功制备了稳定分泌抗人H-FABP单克隆抗体的杂交瘤细胞株。 |

| Abstract: | Objective:Preparition of monoclone antibody to human heart type fatty acid-binding protein.Methods 1)The BALB/C mouse was immuned with intraperitoneal by recombinant humen H-FABP.2)The spleen cells and myeloma cells of the mouse were fused and cultivated,then monoclonal antibody of human H-FABP was selected.3)The antibody was purificated by Protein G Sepharose 4 Fast Flow affinity chromatograph and it was identified.Results 1)Two strains monoclonal antibody which had high affinities and far space sites were gotten and purified.Conclusion The monoclonal antibody of human H-FABP was successfully prepareted. |
