核酶阻断IGF-1及IGF-1R表达抑制病理性瘢痕形成的研究

摘    要:目的:探讨胰岛素样生长因子-1(IGF-1)及其受体(IGF-1R)在瘢痕组织中的表达水平及核酶对其表达的影响作用.方法:选取2015年5月至2016年7月广州红十字会医院烧伤整形科手术切除的18例患者的病理性瘢痕组织(增生瘢痕9例、瘢痕疙瘩9例),同期收集的20例正常皮肤标本组织,采用苏木精-伊红染色对组织进行观察,采用实时荧光定量PCR技术检测各组织标本的IGF-1 mRNA及其受体mRNA的表达水平,取瘢痕组织成纤维细胞进行细胞培养获得IGF-1及其受体核酶,取第2-4代生长状态良好的对数生长期细胞+核酶继续培养72h后采用实时荧光定量PCR技术检测IGF-1 mRNA及其受体mRNA的表达水平.结果:正常皮肤组织、增生性瘢痕组织、瘢痕疙瘩组织中的IGF-1 mRNA及其受体mRNA的表达水平差异具有统计学意义(P<0.05),组间比较呈现正常皮肤组织<增生性瘢痕组织<瘢痕疙瘩组织的特点且差异均具有统计学意义(P<0.05).正常皮肤组织、增生性瘢痕组织、瘢痕疙瘩组织中的IGF-1 mRNA及其受体mRNA的表达水平差异具有统计学意义(P<0.05),组间比较呈现正常皮肤组织<增生性瘢痕组织<瘢痕疙瘩组织的特点且差异均具有统计学意义(P<0.05).结论:瘢痕组织中IGF-1 mRNA及其受体mRNA的表达水平高于正常组织,核酶具有阻断IGF-1 mRNA及其受体mRNA表达的作用,可能在抑制瘢痕的形成中发挥作用.

Effect of Ribozyme on Blockade of IGF-1 and IGF-1R Expression as Well as Inhibition of Scar Formation
Abstract:Objective:To discuss insulin-like growth factor receptor 1 ( IGF-1) and its receptor ( IGF-1R) expression in scar tissue and the effect of ribozyme on the expression level. Methods:18 cases of Patho-logical scar tissue from operation excision (9 cases of scar keloid, 9 cases of scar hyperplasia ) were selected in Department of Burn and Plastics, Guangzhou Red Cross Hospital from 2015 May to 2016 July, meanwhile 20 cases of normal skin tissue specimens were collected. The expression level of tissue was observed by using hematoxylin eosin staining, the expression level of IGF-1 mRNA in each tissue specimen and its receptor mR-NA were detected by using real-time fluorescence quantitative PCR, scar tissue fibroblasts were derived to culture cells to obtained IGF-1 and its receptor ribozyme, the cells in the logarithmic growth phase with good growth state in 2-4 generation + ribozyme were selected to culture,72h later, the expression level of IGF-1 mRNA and its receptor mRNA were detected by using real-time fluorescence quantitative PCR. Results:The differences were statistically significant in the expression level of IGF-1 mRNA in normal skin, hypertrophic scar and keloid tissue and its receptor mRNA ( P <0.05) , the comparison between groups showed that normal skin hypertrophic scar tissue
