枸橼酸芬太尼对乳腺癌细胞体外增殖和侵袭的影响

摘    要:目的:探讨枸橼酸芬太尼对人乳腺癌MCF-7细胞体外增殖和侵袭的影响。方法:分别用0.000 1μmol/L、0.01μmol/L、1μmol/L的枸橼酸芬太尼处理MCF-7细胞,MTT比色法检测MCF-7细胞增殖;流式细胞仪检测MCF-7细胞细胞周期;采用划痕实验及侵袭小室法检测MCF-7细胞侵袭能力。结果:MTT比色法显示不同浓度枸橼酸芬太尼处理的MCF-7细胞的增殖率分别为(58.84±11.31)%、(54.37±9.89)%和(51.83±10.33)%,明显低于对照组;各组MCF-7细胞停滞在G2/M期的比例增加,S期比例减少;各实验组及对照组划痕实验显示48 h时间段的愈合率分别为(70.41±6.86)%、(64.36±3.87)%、(62.52±4.95)%和(83.34±4.59)%;侵袭小室实验显示枸橼酸芬太尼在体外具有抑制MCF-7细胞侵袭转移作用。结论:芬太尼可抑制人乳腺癌MCF-7细胞的增殖、使细胞周期停滞于G2/M期,并且降低其侵袭能力,对MCF-7细胞的生物学性状产生较大的影响。

Effect of Fentanyl citrate on proliferation and invasion of breast cancer cells in vitro

Abstract:Objective:To explore the effect of Fentanyl citrate on proliferation and invasion of breast cancer cell line MCF-7 in vitro.Methods:MCF-7 cells were treated with Fentanyl citrate of different concentrations(0.000 1 μmol/L,0.01 μmol/L,and 1 μmol/L),MTT assay was used to detect the proliferation of MCF-7 cells;flow cytometry was used to detect the cycle of MCF-7 cells;wound healing assay and invasive cabinet method were used to detect the invasive ability of MCF-7 cells.Results:The results of MTT colorimetry showed that the proliferation rates of MCF-7 cells treated with different concentrations of Fentanyl citrate were(58.84±11.31)%,(54.37±9.89)%,and(51.83±10.33)%,respectively,which were significantly lower than those in control group;in different groups,the proportions of MCF-7 cells in G2/M stage increased,while the proportions of MCF-7 cells in S stage decreased;the results of wound healing assay showed that the healing rates at 48 hours in different experimental groups and control group were(70.41±6.86)%,(64.36±3.87)%,(62.52±4.95)%,and(83.34±4.59)%,respectively;the results of invasive cabinet method showed that Fentanyl citrate played an inhibiting role in invasion and metastasis of MCF-7 cells in vitro.Conclusion:Fentanyl can inhibit the proliferation of human breast cancer MCF-7 cells,arrest the cell cycle at G2/M stage,reduce the invasive ability of MCF-7 cells,and it has big impact on biological characteristics of MCF-7 cells.
