IL-12在血吸虫感染小鼠肝脏中的表达及其对肝纤维化的影响

目的探讨日本血吸虫感染小鼠肝脏中白细胞介素-12（IL-12）的表达及其对肝纤维化的影响。方法在小鼠感染血吸虫后的不同时期（6、8、10、12周），分别采用免疫组织化学法观察肝脏IL-12的变化，VG染色及多媒体病理图文定量分析系统检测注射基因重组小鼠IL-12（rIL-12）前、后胶原纤维含量的改变。结果小鼠肝脏IL-12含量随感染时间延长而缓慢下降，胶原纤维含量随感染时间延长逐渐升高，经皮下注射rIL-12后小鼠肝脏IL-12含量随着相应因子补充而显著上升，胶原纤维含量则逐渐降低。结论IL-12能明显抑制血吸虫感染小鼠肝脏胶原纤维合成，延缓肝纤维化发生。

Expression of IL-12 and the Effects of rIL-12 on Liver Fibrosis in Mice Infected with Schistosoma japonicum

Objective To detect the expression of interleukin-12 (IL-12) in the liver of mice infected with Schistosoma japonicum and study the effects of IL-12 on liver fibrosis after supplementary injection of the cytokine.Methods At different time points (6, 8, 10, 12 weeks) after the mice were infected with Schistosoma japonicum, ABC immunohistochemical method was used to dynamically observe the changes in IL-12 in liver. The VG dye and pathologic image multimedia quantification system were used to detect the amount of collagen in the liver tissue of the mice before and after injection of recombinant IL-12 (rIL-12).Results The level of IL-12 in the liver tissue of the mice was gradually decreased with the prolongation of the infection,while the content of collagen was gradually increased (P<0.01). After subcutaneous injection of rIL-12, the levels of IL-12 in the liver tissue of the mice were significantly elevated, while the content of collagen was gradually decreased.Conclusion IL-12 could markedly inhibit the synthesis of collagen in the liver of the mice infected with Schistosoma japonicum and delay the development of liver fibrosis.
