补肾A方对老龄SD大鼠细胞因子表达的调节作用

目的：研究中医补肾药方（A方）对老龄SD大鼠细胞因子表达的影响。方法：把12月龄SD大鼠随机分为实验组和对照组,实验组给予灌服补肾A方药液,90天后处死动物,摘取脾脏,提取RNA。同时采用3月龄的SD大鼠作为年轻对照组。采用美国Affymetrix公司RayBio大鼠细胞因子抗体芯片试剂盒,检测细胞因子表达水平,并进行对照比较。结果：15月龄大鼠比3月龄大鼠的IL-10表达增加,而CNTF、β-NGF、TNF-α、IL-1α、IL-1β、IL-6的表达下调;灌服补肾A方的大鼠比同龄对照组大鼠上调了CINC-3、IFN-α、LIX、TNF-α、VEG和Leptin细胞因子的表达水平。结论：机体细胞因子的表达随着增龄而变化,中医补肾治疗可以调节随增龄而变化的细胞因子表达,这可能是中医补肾抗衰老的机理之一。

Regulation Effect on the Expression of Cytokins in Aged Rat of SD With Recipe A of Restoring Kidney of Traditional Chinese Medicine

Purpose：To investigate effect on the expression of cytokins in aged rat of SD with recipe A of restoring kidney of traditional Chinese medicine（TCM）. Experimental Design：8 of male rats of Sprague dawley（ SD）of 3mo. with clean and healthy were selected as young control group; 16 of male rats with 12mo. with clean and healthy were randomly allocated to experimental group Ⅰ , 8,and 8 as older control group. Recipe A for restoring kidney composed of gouqi, shouwu and yinyanghuo of TCM, was made to the liquor with lg/ml,respectively. Experimental group Ⅰ were given Recipe A liquor in the light of 5g/kg for 90 days. The RNA was extracted from spleen taken out of rats, respectively. The cytokines were determined with Rat Cytokines Antibody Array Ⅰ by Affymetrix of USA. Results： Cytokines in rats of 15mo. are changing with aging, namely elevation of IL- 10 and decline of CNTF,β- NGF,TNF-α, IL- 1α, IL-β and IL-6. The up regulation of cytokines, such as CINC-3, IFN-α, LIX,TNF-α, VEGF and Leptin, after treatment with Recipe A liquor. Conclusions ： Cytokines in aged rats are changing with aging ; restoring kidney of TCM may changed the condition and may be one of mechanisms by TCM.
