IL-10基因启动子区CpG岛甲基化与宫颈癌的相关性研究

目的:探讨IL-10基因启动子甲基化与HPV感染导致宫颈癌的相关性。方法:采用PCR和甲基化特异性PCR(MSP)分别检测宫颈癌49例、癌前病变50例(19例CIN1、10例CIN2、21例CIN3)及正常对照50例宫颈组织中HPV16及IL-10基因启动子甲基化状态,并分析IL-10基因甲基化状态与不同临床病理特征之间的联系。结果:宫颈癌组及癌前病变组IL-10基因甲基化率与对照组比较差异均有统计学意义(P﹤0.05)。宫颈癌组IL-10基因甲基化和HPV16 E7 DNA检测结果有一定的关联性(χ2=20.163,P<0.05,Pearson列联系数r=0.345)。不同年龄、肿瘤组织大小、病理分级、临床分期及有无淋巴结转移和IL-10基因甲基化未见明显关系(P>0.05)。结论:宫颈癌IL-10基因启动子CpG岛甲基化与高危型HPV16感染具有关联性,IL-10基因启动子区低甲基化与宫颈癌发病相关。

Relationship between promoter CpG methylation of IL-10 gene and cervical cancer

Objective:To investigate the promoter methylation status of IL-10 gene in cervical cancer and the correlation with clinicopathologic characteristics.Methods:Promoter methylation was evaluated by using methylation-special polymerase chain reaction(MSP) in 49 cervical cancer tissues specimens,50 cervical intraepithelial neoplasia(CIN: CIN1 n=19,CIN2 n=10,CIN3 n=21) and healthy control(n=50).The relationship between clinicopathologic parameters and methylation status was also evaluated.Results:Compared with control,methylation frequencies in both cervical cancer and CIN were higher(P﹤0.05).IL-10 gene methylation and human papilla virus(HPV) 16 E7 DNA had consistency(χ2=20.163,P<0.05,r=0.345).There were no significant associations between the methylation frequencies for IL-10 and age,tumor size,FIGO stages and WHO grades,lymph node metastasis(P>0.05).Conclusion:Promoter methylation of IL-10 and HPV16 DNA testing has consistency.IL-10 gene promoter methylation has a significant correlation with cervical cancer.
