维甲酸受体β在宫颈癌中的表达及其意义

目的:检测维甲酸受体β(RARβ)基因在宫颈癌组织中的表达及其意义。方法:收集该院68例宫颈癌和28例宫颈炎患者的临床资料和组织标本,RT-PCR法检测宫颈癌和宫颈炎组织中RARβmRNA的表达水平。采用免疫组织化学法和酶联免疫吸附试验检测宫颈癌和宫颈炎组织中RARβ蛋白的表达情况。统计两组临床资料并对随访的患者资料进行对比。结果:宫颈癌组中的RARβmRNA相对表达量低于宫颈炎组,宫颈癌组中的RARβ蛋白表达阳性比例低于宫颈炎组,宫颈癌组中的RARβ蛋白含量平均值低于宫颈炎组平均值,差异均具有统计学意义(P<0.05)。RARβ蛋白在宫颈癌Ⅲ/Ⅳ期阳性表达比例低于Ⅰ/Ⅱ期,差异具有统计学意义(P<0.05)。有淋巴结转移患者中RARβ表达阳性比例低于无淋巴结转移,差异具有统计学意义(P<0.05)。RARβ表达阴性的宫颈癌患者中死亡率和复发率高于阳性表达患者,差异均具有统计学意义(P<0.05)。结论:RARβ在宫颈癌的发生过程中发挥重要作用,故其可作为宫颈癌患者发生、发展和预后判断的重要参考指标。

Expression and significance of retinoic acid receptor-beta in cervical cancer

Objective:To investigate the expression and significance of retinoic acid receptor-beta(RARβ)in cervical cancer.Methods:The clinical data and tissue samples from 68 cases with cervical cancer and 28 cases with cervicitis were collected in our hospital.RT-PCR was used to examine the expression of RARβ mRNA.Immunohistochemistry and ELISA were used to detect the expression RARβ protein.Results:The RARβ mRNA expression rate in cervical cancer was significantly lower than that of cervicitis tissue(P<0.05).The positive RARβ protein expression rate in cervical cancer was significantly lower than that of cervicitis tissue(P<0.05).The RARβ protein content in cervical cancer was significantly lower than that of cervicitis tissue(P<0.05).The positive RARβ protein expression rate in cervical cancer stage Ⅲ / Ⅳ was significantly lower than that of cervical cancer stageⅠ/Ⅱ(P<0.05).The positive RARβ protein expression rate was significantly higher in patients without lymph node metastasis than that with lymph node metastasis(P<0.05).The mortality and recurrence rate in patients with negative RARβ expression were significantly higher than those of patients with positive expression(P<0.05).Conclusion:The defective expression of RARβ gene plays an important role in the carcinogenesis of cervical cancer.Therefore RARβ can be used as an important indicator for the development and prognosis in patients with cervical cancer.
