海尔福对去卵巢和铅中毒大鼠骨组织及血液影响的实验研究

摘    要:目的:观察海尔福对去卵巢和铅中毒大鼠骨组织及血液影响,了解海尔福对去卵巢和铅中毒大鼠骨骨质疏松治疗作用,为雌激素缺乏和铅中毒所引致骨质疏松症的防治提供实验依据。方法:选取5月龄健康雌性大鼠100只,分成5组。对照组、去卵巢模型组(OVX)、铅中毒模型组(LPD)、海尔福+去卵巢模型组(H+OVX)和海尔福+铅中毒模型组(H+LPD)。16周后,麻醉大鼠取左心血,处死后取双侧股骨。股骨切片光镜观察骨组织形态结构,并对血液中的钙、磷、铅、碱性磷酸酶和雌二醇进行测定,数据进行统计学分析。结果:H+LPD组骨组织结构、血钙、血磷和血红蛋白及碱性磷酸酶和雌二醇正常;0VX组、LPD组和H+OVX组的血钙及血磷均明显降低(P﹤0.01),血碱性磷酸酶活力升高(P﹤0.01),骨组织形态呈现骨质疏松的病理改变;0VX组和H+OVX组的雌二醇降低(P﹤0.01);LPD组的血铅明显高于对照组(P﹤0.01)。结论:去卵巢和铅中毒均可引起大鼠骨质疏松,铅中毒影响血红蛋白合成,海尔福对铅中毒所致的大鼠骨质疏松有治疗作用,而对雌激素缺乏大鼠骨质疏松治疗无效。

Experimental study on effects of Haierfu on bone tissue and blood of the ovariectomized or lead poisoning rats
Abstract:Objective:To observe the efficacy of Haierfu in the ovariectomized or lead poisoning rats treatment for osteoporosis by watching the effects of Haierfu on the rats' bone tissue and blood,and provid experimental evidence for the prevention and treatment for osteoporosis caused by estrogen deficiency and lead poisoning.Methods:100 rats of 5-month-old healthy female were selected and divided into five groups: control group,ovariectomized model group(OVX),lead poisoning model group(LPD),Haierfu + ovariectomized model group(H + OVX) and Haierfu + lead poisoning model group(H + LPD).After 16 weeks,the rats were anesthetized to take left heart blood and killed to take bilateral femur.The femur sections were observed morphologically under light microscopy,blood calcium,phosphorus,lead,alkaline phosphatase and estradiol were measured.The data was used for statistical analysis.Results:Structure of bone tissue,calcium,phosphorus,hemoglobin,alkaline phosphatase and estradiol of the rats in H + LPD group were normal;In OVX group,LPD group and H + OVX group,serum calcium and phosphorus were significantly lower(P<0.01),blood alkaline phosphatase activity were increased(P<0.01),and osteoporosis could be found in bone tissue morphologically;In OVX group and H + OVX group estradiol were decreased(P<0.01);In LPD group,blood lead was higher(P<0.01).Conclusion:Ovariectomization and lead poisoning could cause osteoporosis of rats,lead poisoning affect hemoglobin synthesis.Haierfu could have therapeutic effects on osteoporosis of rats caused by lead poisoning,but not by estrogen deficiency.
