拜耳ADVIACentaur测定血清抗甲状腺过氧化物酶抗体浓度的方法学评价

摘    要:目的评价拜耳ADVIA Centaur直接化学发光法测定血清抗甲状腺过氧化物酶抗体(anti—TpoAb);方法用AD—VIA Centaur进行特异性评价实验,功能灵敏度实验,方法学相关性评价实验,稀释回收率实验和加样回收率实验;结果体内生物学活性类似的物质对测试方法无干扰,方法最低的功能灵敏度为25U/ml,直接化学发光法测定anti—TpoAb与经典的放射免疫分析法有良好的相关性,稀释回收实验和加样回收实验结果均满足实验要求;结论拜耳的ADVIACentaur所采用的直接化学发光法是一种先进的检测anti—TpoAb的方法,具有较经典放免分析法简单、快速、灵敏度高,检测速度快、无放射性污染等优点。

Abstract:Objective To evaluate the method of direct chemiluminescence assay to detect antithyroperoxidase antibody(Anti-TPOAb) by bayer ADVIA Centaur.Methods By bayer ADVIA Centaur,specifity valuation test,function sensitivity test,methodology correlation valuation test,dilution recollection rate test and dispense recollection test were carried out.Results Interference of human blood serum with this method is not found.The lowest sensitivity of this method is 25U/ml,the anti-TPOAb determined by direct chemiluminescenceassay is strongly and significantly correlated to the value by classical RIA method,the results of dilution recollection rate test and dispense recollection test meet the laboratory need.Conclusions Direct chemiluminescence assay of BAYER ADVIA Centaur is an advanced and promising method to detect antithyroperoxidase antibody(Anti-TPOAb),compared with classical RIA method,it has many advantages with simpleness,rapidness,high sensitivity and no radiation.
