中药益气化瘀方对体外培养的大鼠颈椎间盘软骨细胞基质金属蛋白酶mRNA表达的影响

摘    要:研究中药益气化瘀方对体外培养的大鼠颈椎间盘软骨细胞基质金属蛋白酶mRNA表达的影响。取健康10周龄SD大鼠70只,体重(200±50)g,雌雄各半,随机分为空白组(E)、芬必得组(D)、中药低剂量组(C)、中药中剂量组(B)、中药高剂量组(A),每组14只。定时、定量胃饲,4天后分别处死取血清,相同条件下制备益气化瘀方含药血清和对照血清,取体外培养的大鼠颈椎间盘软骨细胞进行实验研究,采用RT-PCR法。结果显示,各含药血清组培养的软骨细胞均具有分泌MMP-3、TIMP-1功能,但不具有诱导其异常分泌MMP-3、TIMP-1功能。益气化瘀方组较对照组及芬必得组有下调MMP-3的分泌或表达,上调TIMP-1的分泌或表达。

Abstract:Objective: To study the effects of formula of invigorating Qi and removing blood-stasis on proliferation and metabolism of cultured intervertebral disc chondrocyte.Methods:70 healthy aged 10 week,weight(200±50)g,1/2 female & 1/2 male rats were randomly and equally divided into 5 groups.Group E terms as a normal control group,Group A terms as a Chinese herb group(high dosage),Group B terms as a Chinese herb group(moderate dosage),Group C terms as a Chinese herb group(low dosage),Group D terms as a west medicine group(normal dosage),Group N terms as Iubufan.Each group was respectively fed 4 days.To obtain serum of rats from each group after 4 days.Results:Invigorate Qi and remove blood-stasis group stimulated proliferation and metabolism of chondrocyte at doses ranging from low to high.Maximal effect of stimulation occurred at high dose after treatment(P<0.01).The time of DNA synthesis(S) was faster in experimental group than in control group.Conclusion:Invigorate Qi and remove blood-stasis group can stimulated proliferation and metabolism of chondrocyte in experimental study of cell culture(P>0.05).The time of DNA synthesis(S)was faster in experimental group than in control group.It can play an active role in delaying the degeneration of intervertebral discs.
