骨肉瘤中PTEN蛋白的表达与微血管密度相关性研究

摘    要:【目的】研究骨肉瘤组织中PTEN蛋白的表达与骨肉瘤微血管密度的相关性,为阐明骨肉瘤发生发展的分子机制提供一定的依据。【方法】利用免疫组织化学技术S-P法检测18例骨软骨瘤和36骨肉瘤中的PTEN、CD34蛋白的表达,对其结果进行相关分析。【结果】①PTEN在骨肉瘤中和骨软骨瘤的阳性表达率分别为58.3%和88.9%,骨肉瘤组低于骨软骨瘤组,PTEN蛋白在骨肉瘤和骨软骨瘤组织中的阳性表达率有显著性差异(P〈0.05)。②在骨肉瘤组织中,Ⅲ期骨肉瘤PTEN蛋白的阳性表达率与Ⅱ期(ⅡA期、ⅡB期)组相比明显下降,有显著性差异(P〈0.05),而ⅡA期与ⅡB期骨肉瘤的PTEN蛋白的表达率没有显著性差异(P〉0.05)。③在骨肉瘤组织中,PTEN蛋白的表达与患者的性别、年龄及病理组织学分型未见明显相关性。④骨肉瘤组织中PTEN蛋白表达强度与骨肉瘤微血管密度(MVD)之间呈明显负相关(r=-0.915,P〈0.01)。【结论】①PTEN蛋白在骨肉瘤组织中低表达,在骨软骨瘤组织中高表达;在骨肉瘤的Ennecking外科分期中,ⅡA、ⅡB期的PTEN蛋白表达高于Ⅲ期,PTEN蛋白的表达与骨肉瘤的恶性程度、骨肉瘤的浸润、转移相关。②提示PTEN蛋白在骨肉瘤组织中表达强度与肿瘤微血管密度MVD之间呈明显负相关,骨肉瘤PTEN蛋白表达的缺失和降低可能促进肿瘤血管的生成,在骨肉瘤发生发展过程中起着重要作用。

Relationship Between the Expression of PTEN Proteins and Microvessel Density in Human Osteosarcoma

Abstract:Objective] To study the expression of PTEN protein in human osteosarcoma, and to investigate the relationship between PTEN expression and microvessel density(MVD) in human osteosarcoma so as to provide an evidence for the molecule mechanism of sarcomagenesis and development of osteosarcoma. Methods] Thirty six cases of human osteosarcoma specimen were selected in the study and compared with 18 cases of osteochondroma. The expression of PTEN and CD34 on osteosarcoma was detected by S-P immuno- histochemistry method and the expression relationship was analyzed. Results]The positive expression rate of PTEN in osteosarcoma was 58. 3% which was lower than that (88.9%) of osteochondroma, and there was statistically significant difference between the two groups ( P 〈0.05). The expression of PTEN was negative ly correlated with Ennecking staging of osteosarcoma. The positive rate of stage Ⅱ ( ⅡA, Ⅱ B) was remarkably higher than that of stage Ⅲ , and there was statistically significant difference between the two stages ( P〈 0.05), while the positive rate had no significant difference between stage Ⅱ A and stageⅡ B( P 〉0.05). The positive rate had no significant difference among different age, sex and pathological types ( P 〉0.05). There was negative correlation between the MVD and the expression of PTEN proteins in osteosareoma( r= - 0. 915, P 〈0.01). Conclusion] The protein of PTEN is expressed in osteosareoma at low level and highly expressed in osteochondroma. The expression rate of PTEN in stage Ⅱ A and Ⅱ B is significantly higher than that in stage Ⅲ. The low expression of PTEN is correlated with the malignancy of osteosarcoma and may be involved into the infiltration and metastasis of osteosareoma. There is negative correlation between the MVD and the expression of PTEN proteins in osteosarcoma. Loss of PTEN may be involved into angiogenesis and play a cer rain role in the occurrence and development of osteosarcoma.
