摘    要:目的比较实时荧光聚合酶链反应(PCR)与第2代杂交捕获法(HC-Ⅱ)2种人乳头瘤病毒(HPV)检测方法。方法采用实时PCR与HC-Ⅱ分别检测223例宫颈脱落细胞标本中的高危亚型HPV,对结果不一致的标本进行测序分析。结果实时PCR和HC-Ⅱ2种方法检测高危亚型HPV的一致性较好,总符合率为77%,总一致性指数(Kappa指数)为0.538。其中,高度鳞状上皮细胞内病变(HSIL)组二者的符合率为82%,Kappa指数为0.410,一致性比其他组好。结论实时PCR与HC-Ⅱ检测结果一致性较好,尤其对于HSIL患者,可作为临床上HPV高危亚型检测的有效手段。

Abstract:Objective To compare real-time polymerase chain reaction (PCR) with hybrid capture Ⅱ ( HC-Ⅱ)for detection of Human papillomavirus (HPV). Methods Real-time PCR and HC-Ⅱ were used for detection of high-risk HPV in 223 female genital samples. Discordant results between the two methods were confirmed with DNA sequencing. Results The results of real-time PCR and HC-Ⅱ showed good accordance for detection of high-risk HPV in female genital samples. The total accordant rate of real-time PCR and HC-Ⅱwas 77% and general Kappa index was 0. 538. The accordance of the two methods in high-grade squamous intraepithelial lesion (HSIL) group was much better than in other groups. The accordant rate and Kappa index of HSIL group were 82% and 0. 410 respectively. Conclusions Real-time PCR is an effective method for detection of high-risk HPV infection in cervix, and is in good accordance with HC-Ⅱ, especially for HSIL samples.
