摘    要:目的探讨端粒酶hTR、hTERT基因在人成釉细胞瘤(Ameloblastoma,AB)中的表达,并与p53蛋白相比较,探讨其临床生物学意义。方法用原位杂交及免疫组化技术,检测54例AB的hTR和hTERT的表达及49例AB的p53蛋白,将二者进行分析,同时以7例口腔黏膜为对照。结果94.4%(51/54)的AB有hTR、hTERTmRNA的表达,81.2%(13/16)和87.5%(14/16)的牙源性角化囊肿(Odontogenickeratocyst,OKC)及2/7和1/7例的正常口腔黏膜分别有hTR和hTERTmRNA表达,3组相比差异有显著性(P<0.001)。hTR与hTERT在AB中表达具有相关性(rs=1.000,P<0.001)。p53蛋白在85.7%(42/49)AB、44%(11/25)OKC、33.3%(1/3)正常口腔黏膜上皮的细胞核为阳性。hTERT与p53蛋白的rs=0.754,P<0.001,hTR与p53蛋白的rs=0.536,呈正相关,P<0.001。结论hTR和hTERT在AB中的检出率高于p53蛋白,其间具有一定相关性(P<0.001),并与AB的生物学行为有关。

Abstract:Objective To detect the expression of p53protein,hTR,and hTERT mRNA in ameloblastoma(AB)and to in-vestigate the clinical biological characteristics of AB.Methods We detected hTR and hTERT Mrna in54cases of AB by in situ hybridization,and p53protein by S-P method.Results The positive ratio of hTR,hTERT mRNA was2/7and1/7in normal oral mucosa,81.2%(13/16)and87.4%(14/16)in odontogenic keratocyst(OKC),94.4%(51/54)in AB,respec-tively.There was a significant difference in these three groups(P<0.001).A strong correlation between hTR and hTERT mRNA was found(rs=1.000,P<0.001).p53protein was expressed in87.5%(42/49)AB,44%(11/25)OKC,33.3%(1/3)normal oral mucosa,respectively.A strong correlation between hTERT mRNA and p53protein was found(rs=0.848,P<0.001).A moderate correlation between hTR mRNA and p53protein was found(rs=0.536,P<0.001).Conclusion The positive ratios of hTR and hTERT were higher than p53protein in AB.Telomerase activity and p53expression play an important role in genesis and development of AB.<
