大鼠气道过敏性嗜酸粒细胞性炎症的时间过程

摘    要:目的:研究大鼠过敏性哮喘模型气道炎症随时间的变化过程;方法:观察卵白蛋白(OA)致敏大鼠在1%OA攻击20min后6h、1d、3d、7d、14d时,支气管肺泡灌洗液(BALF)中白细胞总数及分类的变化、细支气管管壁厚度及支气管壁单位面积嗜酸性粒细胞数的变化;结果:致敏大鼠气雾吸入OA后,BALF中白细胞总数及各类白细胞在6h、1d后明显增多,嗜酸性粒细胞在3d~14d仍有增加;肺切片计算机图像分析结果显示在攻击后14d内细支气管管壁增厚,支气管单位面积内渗出嗜酸性粒细胞数在14d内均明显增加;结论:抗原可诱导气道急性非特异性炎症,嗜酸性粒细胞渗出和支气管壁增厚的反应可持续14d。

Abstract:Objective:To investigate the time-course of airway eosinophilic inflammation in the rat model of allergic asthma.Methods:Two weeks after sensitization to ovalbumin (OA) ,the subjects were challenged with1 % OA aerosol for2 0 min.The total count of leukocytes and the number of the different kinds in the bronchoalveolar lavage (BALF) ,the thickening of bronchiole,and the number of eosinophils around the bronchi were observed atdifferenttime after OA challenge.Results:All kinds of leukocytes in the BAL F significantly increased6 hours and1 day later,and then recovered to normal levels except eosinophils which remained higher level for at least 1 4days after challenge.The imaging analyses of lung sections showed that bronchiole wall were gradually thickened,especially at7and1 4days after challenge,and the number of eosinophils around the bronchi increased markedly throughout the period of1 4days.Conclusion: Antigen induces acute airway inflammation and a long -lasting reaction of eosinophilic inflammation in the lungs of the sensitized rats.
