晶状体损伤对视神经损伤后再生影响的研究

摘    要:目的观察单纯视神经损伤及视神经损伤联合晶状体损伤后视神经中Nogo-A mRNA及Nogo-A的表达,探讨晶状体损伤促进视神经损伤后再生的机制。方法 66只雄性Wistar大鼠随机分为正常对照组(A组,6只)、单纯视神经损伤组(B组,30只)和视神经损伤联合晶状体损伤组(C组,30只)。分别于造模后7d、14d、21d处死大鼠2只(A组)、10只(B组)、10只(C组),光镜下观察视神经的病理变化,免疫组织化学染色检测Nogo-A表达情况,采用逆转录-聚合酶链反应(RT-PCR)、半定量分析不同组视神经Nogo-A mRNA的表达。结果大鼠正常视神经中表达Nogo-A mRNA及Nogo-A,但表达量较低;损伤视神经后7d Nogo-A mRNA的表达量显著升高,至伤后21d B组及C组均维持较高的水平。B组损伤后7d Nogo-A表达阳性的细胞数开始增多(136.80±3.94),与A组比较差异有统计学意义(P<0.05),并保持高表达;C组损伤后21d Nogo-A的表达较B组低,差异有统计学意义(P<0.05)。结论视神经损伤后髓鞘相关抑制因子Nogo-A表达增高,晶状体损伤促进视神经再生的机制可能与Nogo-A有关。

Abstract:Objective To observe the expressive varieties of Nogo-A mRNA and Nogo-A in simple optic nerve injury and optic nerves combined with lens injury,and explore the mechanisms of lens injury promoting optic nerve regeneration.Methods Sixty-six male Wistar rats were randomly divided into normal control group(6 rats,group A),simple optic nerve injury group(30 rats,group B) and joint injury group(30 rats,group C).Two cases in group A,10 cases in group B,10 cases in group C were sacrificed at 7 days,14 days and 21 days after modeling.Pathological changes of optic nerve were observed by optical microscope.The expression of Nogo-A in optic nerve was detected by immunohistochemistry.Semi-quantitative RT-PCR was used to study the expression of Nogo-A mRNA in optic nerve of three groups.Results The normal optic nerve of rats had the expression of Nogo-A mRNA and Nogo-A,but its expression quantity was lower;The expression of Nogo-A mRNA at 7 days after optic nerve injury was significantly increased,and which at 21 days still maintained high level in group B and C.The positive cells which expressed Nogo-A at 7 days after optic nerve injury in group B started to increase(136.80±3.94) and kept high lever,and there was statistical difference compared with the normal group(P<0.05);But the quantity at 21 days after optic nerve injury in group C was lower than that in group B,there was statistical difference(P<0.05).Conclusion After optic nerve injury,the expression of myelin associated inhibitors factor of Nogo-A increases,and the mechanism of lens injury promoting the optic nerve regeneration may be related with Nogo-A.
