摘    要:目的 探讨黄芪注射液对大鼠脑缺血再灌注后血脑屏障的保护作用及ZO-1蛋白表达的影响.方法 采用线栓法制备大鼠局灶性脑缺血再灌注模型.将SD大鼠随机等分为假手术组、生理盐水对照组、黄芪注射液治疗组.采用分光光度计法及免疫组化法分别检测大鼠脑组织伊文氏蓝含量及ZO-1蛋白的表达.结果 与假手术组相比,生理盐水对照组大鼠脑组织伊文氏蓝含量明显增多,ZO-1蛋白的表达明显减少;与生理盐水对照组相比,黄芪注射液组大鼠脑组织伊文氏蓝含量显著减少,ZO-1蛋白的表达湿著增加.结论 黄芪注射液对脑缺血再灌注后血脑屏障具有保护作用,可能与黄芪注射液上调ZO-1蛋白的表达有关.

Abstract:Objective:To investigate the effects of astragus injection on blood-brain barrier and expression of ZO-1 protein after cerebral ischemia and reperfusion.Methods: The string inserting method was employed to make the model of focal cerebral ischemia.SD rats were divided randomly into the sham-operated group,the normal saline control group and the astragus injection group.Evans Blue(EB) leakage and expression of ZO-1 protein were respectively detected by spectrophotometry and immunohistochemistry.Results: Compared with the sham-operated group,EB leakage in brain tissue was increased significantly.The expression of ZO-1 protein was decreased obviously in the normal saline control group.Compared with the normal saline control group,the EB leakage was decreased remarkably and the expression of ZO-1 protein was increased significantly in the astragus injection group.Conclusion: Astragus injection could have protective effect in the blood-brain barrier after cerebral ischemia and reperfusion.Its mechanisms may be associated with up-regulation expression of ZO-1 protein.
