益母草碱对大鼠急性心肌缺血损伤血管舒缩功能及抗氧化作用的影响

摘    要:目的:观察益母草碱对大鼠急性心肌缺血损伤血管舒缩功能及抗氧化作用的影响。方法:垂体后叶素腹腔注射诱导大鼠急性心肌缺血,放射免疫法测定血浆及心肌内皮素(ET)含量,硝酸还原酶法测定血清和心肌一氧化氮(NO)含量,黄嘌呤氧化酶法测定血清和心肌超氧化物歧化酶(SOD)活力,硫代巴比妥酸法测定血清和心肌丙二醛(MDA)含量。结果:益母草碱在一定剂量范围内能升高血中和心肌NO水平(P<0.01),降低ET含量(P<0.01),增加SOD活力和降低MDA含量(P<0.01),且存在量效关系。结论:益母草碱具有对抗垂体后叶素性急性心肌缺血损伤的作用,其机制可能与平衡血管舒缩功能、抗脂质过氧化等有关。

Abstract:Objective:To investigate the equilibrium of vasomotor function and the prevention of lipid peroxidation of leonurine(Leo) on pituitary(Pit)-induced acute myocardial ischemia injury in rats.Methods: Pit was administrated intraperitoneally to induce acute myocardial ischemia injury in rats.The concentration of endothelin(ET) in plasma and myocardium were measured by radio immunoassay.The level of nitric oxide(NO),the activity of superoxide dismutase(SOD) and the content of malondialdehyde(MDA) in serum and myocardium was determined by nitrate reductase assay,xanthine oxidase assay and thibabituric acid assay,respectively.Results: Within certain dose range of Leo,the contents of ET and MDA were obviously decreased and the level of NO and the activity of SOD were increased.Linear regression analysis(scatterplot) indicated that there was dose-effect relationship of Leo against myocardial ischemia injury within certain dose range.Conclusions:Leo inhibits Pit-induced myocardial ischemia injury.The mechanism may be related to the equilibrium of vasomotor function and the prevention of lipid peroxidation.
