鬼针草提取物对实验性高脂血症大鼠血脂和NO及NOS的影响

摘    要:目的研究鬼针草不同提取物对实验性高脂血症大鼠血脂和NO及NOS的影响。方法健康雄性SD大鼠随机分为5组:①正常对照组;②模型对照组;③洛伐他汀对照组,洛伐他汀2mg/kg·d)灌胃;④鬼针草乙酸乙酯萃取物组,5g/(kg·d)灌胃;⑤鬼针草水提取物组,5g/(kg·d)灌胃,正常组和模型组每日给予等体积生理盐水。除正常组喂饲普通饲料外,其余各组均喂饲高脂饲料。10周末处死动物,分离血清,检测血脂、MDA、SOD、NO、NOS。制备腹主动脉切片,HE染色观察形态学改变。结果与模型组比较,鬼针草提取物组TC、LDL—C、MDA、NO和NOS显著降低,HDL—C、SOD明显升高。鬼针草提取物组HDL-C明显高于洛伐他汀组。模型组大鼠主动脉血管内壁不光滑,血管内膜脂质沉积,鬼针草提取物组血管内膜无明显的脂质沉积。结论鬼针草提取物能够降低高脂血症大鼠血脂水平并保护血管内皮,具有较好的防治动脉粥样硬化的作用。

Effects of The Different Extracts of Bidens Bipinnata L.on Blood Lipids,NO and NOS in Experimental Hyperlipidemia Rats

Abstract:Objective To study the effects of the different extracts of Bidens Bipinnata L. on blood lipids, NO and NOS in experimental hyperlipidemia rats. Methods 50 male SD rats selected were randomly divided into five groups :①normal control group ;②model group ; ③Lovastatin group ; ④EtOAc extract of Bidens Bipinnata L.. group; ⑤water extract of Bidens Bipinnata L. group. The rats in each group were feeded with highfat forage except for normal control group, Meanwhile, the rats in normal control group were treated by intragastric administration with isotonic NS, in other drug groups with lovastatin( 2 mg/kg), EtOAc extract of Bidens Bipinnata L. (5 ml/kg) ,water extract of Bidens Bipinnata L. (5 ml/kg) respectively. At the end of the tenth week,All rats were killed, determine the level of biochemical index of blood lipids, MDA, SOD, NO, NOS. abdominal aorta were collected and morphological changes were observed. Results Compared with model group, TC, LDL-C, MDA, NO and NOS was significantly decreased while HDL-C and SOD were increased in the extract of Bidens Bipinuata L. groups. The level of HDL-C was much higher in the extract of Bidens Bipinnata L. than Lovastatin group. Observe the morphology characteristics( HE staining,×400), the inner vessel wall of arteriae aorta of the rats seemed not smooth and fatty substance deposited in the endangium in model group while fatty substance didn' t obviously deposit in the endangium in Bidens Bipinnata L groups. Conclusion All the exacts of Bidens Bipinnata L. could modulate lipid metabolism, to protect vascular endothelium and prevent atherusclerotic.
