白介素-17、白介素-4、干扰素-γ在小儿支气管哮喘诱导痰液中的表达及意义

摘    要:目的探讨白介素-17(IL-17)、白介素-4(IL-4)、干扰素-γ(IFN-γ)在小儿支气管哮喘诱导痰液中的表达及其意义。方法研究对象分支气管哮喘急性发作期组61例和缓解组61例,自身对照,选同期健康体检儿童45例为正常对照组。常规痰涂片染色进行炎性细胞计数并分类,酶联免疫吸附法(ELISA)检测IL-17、IL-4、IFN-γ在诱导痰上清液中浓度。结果IL-17、中性粒细胞计数在急性发作期组明显高于缓解期组,缓解期组高于正常对照组(P〈0.05);IL-4、噬酸性粒细胞计数在急性发作期组高于缓解期组(P〈0.01),缓解期组与正常对照组差异无统计学意义;IFN-γ在急性发作期组低于缓解期组(P〈0.05),缓解期组与正常对照组差异无统计学意义;急性发作期组IFN-γ与IL-4、ILl7呈负相关(r=-0.51,P〈0.05),IL-17与IL-4呈正相关(r=0.58,P〈0,05),缓解期则无相关性。结论IL-17、IL-4参与小儿支气管哮喘的发生和发展,IFN-γ对哮喘的气道炎症有抑制作用,IL-17能更准确的反映支气管哮喘患儿的慢期气道炎症状态。

Abstract:Objective To investigate the production of IL-17,IL-4 and IFN-γ in sputum of pediatric patients with asthma.Methods 61 pediatric patients with asthma in stage of exacerbation were enrolled as exacerbation group.and these patients in remission stage after therapy were enrolled as remission group, and 45 healthy children were chose as control group.The percentage of neutrophils and eosinophils were counted in regular way with sputum pigmentation,and the level of IL-17,IL-4 and IFN-γ were detected by enzyme-linked immunosorbent assay.Results IL-17 in exacerbation group Was highest among three groups, and IL-17 in remission group was higher than that in control group.In exacerbation group,IL-4 was higher and IFN-γ was lower than those in other two groups,and no significant difference Was found between remission group and control group.While in exacerbation stage,IL-17 was positive correlated with IL-4,and IFN-γ was negative correlated with IL-17 and IL-4.Conclusions IL-17 and IL-4 are related to the occurrence and exasperating of asthma,and IL-17 is suit for indicating the inflammation of asthma.IFN-γ is a protective factor of asthma.
