产前诊断中羊水细胞培养方法的研究

目的:探讨产前诊断中羊水细胞培养方法及不同羊水量对培养结果的影响。方法:155例羊水标本,以不同羊水量分为两组,分别以平皿盖玻片法和培养瓶法进行培养,比较两种培养方及不同羊水量的培养结果。结果:平皿盖玻片联合培养瓶法培养一次成功并获满意核型151例,成功率97.4%;两种培养方法比较,两组中均以平皿盖玻片法成功率高于培养瓶法;平皿盖玻片法6~8mL与15~20mL羊水的培养成功率无显著差异。结论:羊水细胞培养中,平皿盖玻片法优于培养瓶法,前者具有成功率高、收获时间早、容易区分真假嵌合型的优点,更适用于羊水量少、孕周偏大者;产前诊断中推荐以培养瓶法联合平皿盖玻片法行羊水细胞培养。

To Study on the Methods of Amniotic Fluid Culture in Prenatal Diagnosis

Objective:The aim of this study is to explore the result of amniotic fluid cell culture in different methods and different volume.Methods:Tolally 155 samples were cultured with in coverslip dishes and flasks that divided into two groups according the volume of amniotic fluid.Results:The general successful rate was 97.4% with coverslip dish combine flask culture.It has high successful rate in coverslip dish culture than in flask culture in two groups.There was no difference between 15~20 mL and 6~8 mL amniotic fluid in coverslip dish culture.Conclusion:In coverslip dish culture is better than in flask culture in amniotic fluid cell culture.It has the advantaggs about the high successful rate,short culture period,easly to distinguish true mosaicism and especially in less volume amnictio fluid and late gestational week.Coverslip dish culture combine with flask culture is suggested in amniotic fluid cell culture in prenatal diagnosis.
