顺铂联合5-氟尿嘧啶对肺癌A549细胞生长的影响

摘    要:目的揍讨顺铂和5-氟尿嘧啶(5-Fu)在体外联合用药对人肺癌A549细胞的生长抑制和促凋亡作用。方法运用四甲基偶氮唑盐(MTT)法检测顺铂和5-FU单独及共同作用48h后对A549细胞生长的影响,每个药物浓度设6个复孔,实验重复3次,结果取平均值;并用流式细胞术检测两药合用对细胞周期和凋亡的影响。结果MTT法结果显示顺铂和5"FU联合使用对A549细胞的生长抑制率高于两药单独使用(P〈0.01);流式细胞仪检测可见两药合用后晚期凋亡明显增多,S期细胞增多(P〈0.01)。结论在体外实验中,顺铂与5-FU联合使用能够更好地抑制肺癌A549细胞的增殖,并可改变细胞周期分布,促进细胞凋亡。

Abstract:Objective To explore the proliferation inhibition and proapoptosis effects of cisplatin combined 5-fluorouracil on human non-small cell lung cancer A549 cells in vitro.Methods The absorbance value was tested by colorimetry through MTT method to evaluate the effect of cisplatin and 5-fluorouracil after 48 hours.Each drug concentration was in six holes and it repeated three times.The influence of cell cycle and apoptosis of cisplatin and 5-fluorouracil was used flow cytometry detection.Results It was proved that cisplatin and 5-fluorouracil could inhibit the proliferation of A549 cells.The inhibition was higher than they alone and it had statistically significant difference(P < 0.01).The cell cycle analysis showed that the percentage of S phase cells increased.The cell apoptosis analysis showed that the percentage of late apoptosis increased(P < 0.01).Conclusion It can be concluded that cisplatin and 5-fluorouracil could inhibit the proliferation of A549 cells in vitro.This inhibition enhances when cisplatin is combined with 5-fluorouracil.
