硝酸甘油抗B组柯萨奇病毒的作用特点和机制

摘    要:目的 研究硝酸甘油(GTN)抗柯萨奇B组3型病毒(CVB3)的作用特点和机制。方法 比较先经GTN预处理及与GTN同时加入的两种处理方法相同滴度的病毒所引起的CPE抑制率的差异;分别测定并比较多次给予GTN及不同时间点给予GTN对CVB3感染的HeLa细胞组间的细胞病变效应抑制效果;测定GTN在HeLa细胞中不同时间点代谢产生的N0浓度,并做N0浓度与不同时间点给予GTN的细胞病变效应抑制结果的相关性分析。结果 预加GTN组与同时加GTN组经等量病毒攻击所引起的CPE无显著性差异(P〉0.05);多次给药的组间CPE抑制率无显著性差异(P〉0.05),但不同时间点给药的组间CPE抑制率具有显著差异(P〈0.05),NO浓度与不同时间点给予GTN的细胞病变效应抑制结果呈正相关(r=0.97,P〈0.01)。结论 GTN是通过其代谢产物NO来发挥抗CVB3作用的。

Abstract:Objective To investigate the antiviral effect and the related mechanism of glyceryl trinatrate(GTN).Methods The cytopathic effect(CPE) inhibitory rate between two groups that were treated with equal amount of CVB3 was compared,and one group was treated with GTN before CVB3 attack while another group was administrated with GTN and CVB3 simultaneously.The CPE inhibitory rate caused by GTN were ex-amined and compared.The nitric oxide(NO) concentrations produced from GTN in HeLa cell were also deter-mined,the correlation between NO concerntrations and CPE inhibitory rates caused by different GTN adminis-trations was analyzed.Results There was no significantly difference between the inhibitory rates in cells treat-ed with GTN before CVB3 attack and with GTN and CVB3 simultaneously(P>0.05).The same result was obtained in the comparision of cytopathic effect inhibitory rate given GTN at different time points(P>0.05).The difference of CPE inhibitory rate between the two groups was significant(P<0.05).NO concentrations were highly correlated with CPE inhibitory rate(r=0.947,P<0.01).Conclusion GTN could inhibit CVB3 replicationin vitrovia NO.
