摘    要:目的了解线粒体DNA突变在肿瘤发生发展中的作用.方法应用PCR-限制性片段长度多态性分析技术对LA795、CT26和SCC891等3个肿瘤细胞系mtDNA编码区基因片段进行了分析.结果 mtDNA编码区的tRNAMet Glu Ⅰle及NDI基因核酸片段经HaeⅢ等16种内切酶消化后,这些肿瘤细胞系表现出完全相同的酶切图谱,其酶切格局及酶切片段大小完全相同.结论本组肿瘤细胞系mtDNA编码区基因结构相对稳定.

Abstract:Objective To inquire into the relationship between mutations of mtDNA and carcinogenesis.Methods The variations of ND3 and tRNA Met Glu Ile gene fragments of mtDNA from three tumor cell lines of mice was analyzed by PCR-RFLP techniques.Results After being cleavaging by 16 restriction endonucleases,all restriction patterns of ND3 and tRNA Met Glu Ile gene fragments were same in these tumors.Conclusion The gene structure of coding region of mitochondrial DNA from these tumors is comparatively conserved.
