摘    要:目的:应用不同饮食喂养大鼠,观察大鼠产生胰岛素抵抗(IR)的情况,同时观察大鼠游离脂肪酸(FFA)的变化。方法:分别以普通饲料、高糖饲料、高脂饲料、高糖高脂饲料喂养大鼠8周后,行高胰岛素-正葡萄糖钳夹实验,对大鼠胰岛素抵抗程度进行评估,应用酶联免疫吸附方法检测大鼠血清中游离脂肪酸,测量大鼠体重、附睾脂肪垫重量。结果:高脂饲料、高糖高脂饲料组大鼠产生胰岛素抵抗,普通饲料、高糖饲料组未出现胰岛素抵抗,胰岛素抵抗组大鼠游离脂肪酸明显高于非胰岛素抵抗大鼠,胰岛素抵抗组大鼠体重及附睾脂肪垫重量明显高于非胰岛素抵抗大鼠。结论:高脂饲料、高糖高脂饲料喂养大鼠8周可成功地诱导大鼠产生胰岛素抵抗,胰岛素抵抗的产生可能与肥胖、高游离脂肪酸有关。

Abstract:Objects:To investigate the insulin resistance induced by different diet in rats,and to detect the free fat acid(FFA).Methods:After eight weeks feeding of commondiet,high sugar diet,high fat diet,or high sugar high fat diet,theinsulin resistance of Wistar male rats was evaluated by hyperinsulinism-nor blood sugar ring clamp experiment,the FFA were detected by enzyme linked immunosorbent assay,and the body weight and epididymis fat pad weights were also detected.Results:Insulin resistance was induced in the rats fed by high fat diet and high sugarhigh fat diet,not in the rats fed by common diet nor high sugar diet.The FFA were obviously higher in the insulin resistance rats than in the rats without insulin resistance.body weight especially the weight of epididymal fat pad were obviously higher in the insulin resistance rats than in the rats without insulin resistance.Conclusions:Insulin resistance could be successfully induced by 8 weeks feeding of high fat diet or high sugar high fat diet.The occurence of insulin resistance may be related to fat,high-level FFA,and inflammatiory reaction.
