| 摘 要： | 目的：探讨采用Chelex-100法从颊粘膜拭子提取DNA这一方法的有效性和可靠性。方法：用棉签刮取130例个体口腔颊粘膜脱落细胞，采用Chelex-100法从颊粘膜拭子中提取DNA，采用SSP—PCR及PCR—PFL，P法对其进行检测，观察结果的有效性和可靠性。结果：所有样本中都获得成功。电泳结果显示，PCR扩增及酶切的靶带清晰，无非特异性杂带，扩增结果重复性好。结论：采用Chelex-100法从颊粘膜拭子提取DNA，方法稳定可靠，可以用于基因型检测。 |

| Abstract: | Objective: To investigate the specificity and reliable for extracting DNA from buccal swab by the Chelex 100 method. Methods: The buccal swabs were collected from 130 individuals, Genomic DNA was obtained from these buccal swabs using the Chelex 100 procedure. Genotypes were determined by sequence specific primers polymerase chain reaction (SSP PCR) and PCR restriction fragment length polymorphism(PCR RFLP). Results: The target DNA bands were very clear and no nonspecific bands could be detected. Conclusion: DNA obtained from buccal swabs through Chelex 100 procedure is well suited for analysis of gene polymorphisms. |
