血栓弹力图评价脑创伤大鼠血液凝固状态的价值

摘    要:目的探讨血栓弹力图评价脑创伤大鼠血液凝固状态的价值。方法成年健康雄性SD大鼠24只随机平分为三组:假手术组、模型组与治疗组,每组8只。假手术组不建立脑创伤模型,模型组与治疗组均采用液压打击仪建立脑创伤模型,造模成功后即刻从尾静脉给药,模型组和假手术组给予相同体积的生理盐水,治疗组给予牛磺酸200mg/kg,每组动物连续给药7 d。采用血栓弹力图观察三组反应时间(R)、形成时间(K)、最大切应力系数(MA)值与血栓到达最大幅度后60 min时血栓的纤溶率(LY60)值。采用全自动凝血分析仪检测三组血浆凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)、纤维蛋白原(FIB)。直线相关分析常规凝血指标与血栓弹力图各指标的相关性。结果模型组与治疗组的R值与K值均显著高于假手术组,MA值与LY60值显著低于假手术组,差异有统计学意义(P<0.05),与模型组比较,治疗组R值与K值均显著降低(P<0.05),MA值与LY60值均显著升高(P<0.05)。模型组的FIB值显著高于假手术组(P<0.05),治疗组与假手术组比较差异无统计学意义(P>0.05),三组PT、APTT、TT值比较差异无统计学意义(P>0.05)。在脑创伤大鼠中,直线相关分析显示FIB值与血栓弹力图R值、K值呈显著正相关性(P<0.05),与MA值与LY60值呈显著负相关性(P<0.05)。结论血栓弹力图作为一种凝血状态研究的手段,可与常规凝血指标检测优势互补,用于评价脑创伤大鼠血液凝固状态,有很好的应用价值。

Value of thromboelastography in evaluating blood coagulation status in rats with brain trauma

Abstract:Objective To investigate the values of thromboelastography in evaluating blood coagulation status in rats with brain trauma.Methods 24 adult healthy male SD rats were randomly divided into three groups:the sham operation group,the model control group and the treatment group,with 8 rats in each group.The sham operation group did not establish the brain trauma model.The model group and the treatment group were used a hydraulic impact device to establish brain trauma model.Immediately after successful modeling,the treatment group was administered with taurine of 200 mg/kg through tail vein,while the model group were given the same volume of normal saline,and each group of animals were administered continuously for 7 days.Thromboelastography was used to observe the response time(R),formation time(K),maximum amplitude(MA)value and fibrinolytic rate(LY60)value of the thrombus at 60 minutes after the thrombus reached the maximum amplitude.The plasma prothrombin time(PT),activated partial thromboplastin time(APTT),thrombin time(TT),and fibrinogen(FIB)of three groups were measured using an automatic coagulation analyzer.Linear correlation analysis was used to analyze the correlation between conventional coagulation indicators and various indicators of thromboelastography.Results The R and K values of the model group and the treatment group were significantly higher than those of the control group,the MA value and LY60 value were significantly lower than those of the sham operation group,and the difference was statistically significant(P<0.05).Compared with the model group,the R and K values of the treatment group were significantly lower(P<0.05).,and the MA value and LY60 value were significantly higher(P<0.05).The FIB value of the model control group was significantly higher than that of the control group(P<0.05),and there was no significant difference between the treatment group and the control group(P>0.05).There were no significant differences in the PT,APTT and TT values among the three groups(P>0.05).In the rats with brain trauma,the linear correlation analysis showed that the FIB value were significantly positively correlated with the R and K values of the thromboelastogram(P<0.05),and were significantly negatively correlated with the MA value and LY60 value(P<0.05).Conclusion As a method of studying blood coagulation status,thromboelastography can complement the advantages of conventional coagulation indicators,and can evaluate the blood coagulation state of rats with brain trauma,so it has good application values.
