Satraplatin联合放疗对鼻咽癌裸鼠STAT3及NF-κB表达的影响

摘    要:目的观察并分析Satraplatin(赛特铂)联合放疗对鼻咽癌裸鼠信号传导及转录激活因子(STAT3)及核因子κB(NF-κB)表达的影响。方法取健康裸鼠120只,按照随机数字表法将其分为对照组和观察组,每组60只。所有裸鼠均鼻咽癌造模成功(经过低分化鼻咽癌CNE-2细胞接种)。对照组裸鼠又随机分为未处理组、处理后1周组(放疗)、处理后3周组(放疗),每组20只。观察组裸鼠又随机分为未处理组、处理后1周组(放疗+Satraplatin)、处理后3周组(放疗+Satraplatin),每组20只。观察两组裸鼠处理前、处理后1周、处理后3周肿瘤凋亡率、瘤体重量、STAT3、NF-κB相对表达量。结果两组裸鼠处理前后肿瘤凋亡率差异显著(P <0. 05),两组裸鼠处理前肿瘤凋亡率比较无显著差异(P>0. 05),随着处理后时间的延长,两组裸鼠肿瘤凋亡率显著升高,但观察组裸鼠处理1周、3周肿瘤凋亡率显著高于对照组(P <0. 05)。两组裸鼠处理前后瘤体重量差异显著(P <0. 05),两组裸鼠处理前瘤体重量比较无显著差异(P>0. 05),处理后,两组裸鼠瘤体重量显著降低(P <0. 05),处理1周、3周瘤体重量显著低于对照组(P<0. 05)。两组裸鼠处理前后STAT3、NF-κB水平差异显著(P <0. 05),两组裸鼠处理前STAT3、NF-κB水平比较无显著差异(P>0. 05),处理后,两组裸鼠STAT3、NF-κB水平显著降低(P <0. 05),处理1周、3周观察组裸鼠STAT3、NF-κB水平明显低于对照组(P <0. 05)。结论 Satraplatin联合放疗能有效降低鼻咽癌裸鼠STAT3及NF-κB表达,增加鼻咽癌裸鼠肿瘤细胞凋亡,减轻瘤体重量。

Effects of Satraplatin combined with radiotherapy on STAT3 and NF-kappa B expression in nude mice with nasopharyngeal carcinoma

Abstract:Objective To investigate and analyze the effects of Satraplatin combined with radiotherapy on the expression of STAT3 and NF-kappa B in nude mice with nasopharyngeal carcinoma. Methods 120 healthy nude mice were selected and divided into the control group and the observation group according to random number table method,60 mice in each group. All nude mice were successfully modeled for nasopharyngeal carcinoma( inoculated with poorly differentiated nasopharyngeal carcinoma CNE-2 cells). Nude mice in the control group were randomly divided into untreated group,one-week post-treatment group( radiotherapy),and three-week post-treatment group( radiotherapy),20 mice in each group. The nude mice in the observation group were randomly divided into the untreated group,one-week post-treatment group( radiotherapy + Satraplatin),and three-week post-treatment group( radiotherapy + Satraplatin),20 mice in each group. The tumor apoptotic rate,tumor weight,STAT3 and NF-B relative expression were observed before treatment,1 week and 3 weeks after the treatment in nude mice of the two groups. Results The tumor apoptosis rate of nude mice before and after treatment was significantly different in the two groups( P < 0. 05).There was no significant difference between the two groups before treatment( P > 0. 05). After treatment,the tumor apoptosis rates of the two groups increased significantly,and the tumor apoptosis rates of the observation group was significantly higher than that of the control group at 1 and3 weeks after treatment( P < 0. 05). The tumor weight of nude mice before and after treatment was significantly different in the two groups( P <0. 05). There was no significant difference in tumor weight between the two groups before treatment( P > 0. 05). After treatment,the tumor weight of the two groups decreased significantly( P < 0. 05),and the tumor weight of the observation group was significantly lower than that of the control group at 1 and 3 weeks after treatment( P < 0. 05). The level of STAT3,NF-κB before and after treatment was significantly different in the two groups( P < 0. 05). There was no significant difference in STAT3,NF-κB level between the two groups before treatment( P > 0. 05).After treatment,the STAT3,NF-κB level of the two groups decreased significantly( P < 0. 05). The STAT3,NF-κB level of the observation group was significantly lower than that of the control group at 1 and 3 weeks after treatment( P < 0. 05). Conclusion Satraplatin combined with radiotherapy can effectively reduce the expression of STAT3 and NF-κB in nude mice with nasopharyngeal carcinoma,increase the apoptosis of tumor cells,and reduce the tumor weight.
