丙酮酸乙酯对小鼠胎盘和胎鼠早期生长的影响

摘    要:目的分析丙酮酸乙酯对小鼠胎盘和胎鼠早期生长的影响。方法40只孕鼠随机分为对照组、低剂量组、中剂量组和高剂量组,每组10只。在胎鼠器官形成期即妊娠第7~16天通过腹腔注射方式给药,低、中、高剂量组丙酮酸乙酯注射剂量分别为25 mg·kg^-1·d^-1、50 mg·kg^-1·d^-1和100 mg·kg^-1·d^-1。对照组注射等量生理盐水。在孕鼠妊娠的第17天经眼球取血后处死,通过对孕鼠的行为能力、脏器系数、黄体数、着床率、雌二醇、胎盘组织结构、胎鼠体重及胎鼠畸形等进行分析、检测,探讨丙酮酸乙酯对小鼠胎盘和胎鼠早期生长的影响。结果与对照组比较,胚胎发育期给孕鼠腹腔注射丙酮酸乙酯,中低剂量有助于促进卵巢、胎盘发育,提高胚胎着床率,促进雌二醇表达;过高浓度处理,则会造成卵巢和胎盘的功能的抑制作用。胚胎发育期给孕鼠腹腔注射丙酮酸乙酯,中低剂量有助于促进胚胎成活,降低胎鼠畸形率。结论合适剂量(50 mg·kg^-1·d^-1)的丙酮酸乙酯处理,有助于小鼠胎盘和胎鼠早期生长均有促进作用,过高剂量处理将发生一定的抑制作用。

Abstract:Objective To analyze the effect of ethyl pyruvate on the early growth of mouse placenta and fetus.Methods 40 pregnant rats were randomly divided into control group,low dose group,middle dose group and high dose group,10 rats in each group.During the organogenesis period,from the 7th to the 16th day of pregnancy,the drug was injected intraperitoneally.The doses of ethyl pyruvate were 25 mg·kg^-1·d^-1,50 mg·kg^-1·d^-1 and 100 mg·kg^-1·d^-1,respectively.The control group was injected with the same amount of normal saline.On the 17th day of pregnancy,blood was collected from the eyeball of pregnant mice and then killed.The effects of ethyl pyruvate on the growth of placenta and early fetal mice were studied by analyzing the behavior,organ coefficient,number of corpus luteum,implantation rate,estradiol,placenta tissue structure,fetal weight and fetal deformity of pregnant mice.Results The pregnant rats were injected with ethyl pyruvate intraperitoneally during the development of embryo.The low and medium dose of ethyl pyruvate was helpful to promote the development of ovary and placenta,increase the rate of embryo implantation,and promote the expression of estradiol.The high concentration of ethyl pyruvate could inhibit the function of ovary and placenta.The pregnant mice were injected with ethyl pyruvate intraperitoneally during the development of embryo.The low and medium dose of ethyl pyruvate was helpful to promote the survival of embryo and reduce the rate of malformation.Conclusion Appropriate dose of ethyl pyruvate treatment can promote the early growth of mouse placenta and fetus,and the excessive dose treatment will have a certain inhibitory effect.
