Bcl—2蛋白与胃癌相关性的研究

摘    要:目的:探讨Bcl-2蛋白在胃癌发展过程中的表达和作用。方法:对50例临床病理诊断的胃癌患者标本及10例正常胃粘膜标本进行Bcl-2表达蛋白的免疫组织化学检测,并结合胃癌的分级及癌前病变等分类比较。结果Bcl-2的表达在进展期胃癌(68.0%)、早期胃癌(64.0%)、不同共型性增生(59.0%)、肠化生(57.9%)中均高于正常胃粘膜(20.0%);按胃癌的分化程度;高分化腺癌(82.4%)>中分化腺癌(50.0%)>低分化腺癌(17.6%),并且在癌旁组织中可见增殖带拉长,Bcl-2蛋白在此增殖细胞及周围增生腺体中的表达为强阳性。结论:灵重要的癌基因,在肿瘤发生中起重要作用,胃癌组织中Bcl-2蛋白的表达与组织分化程度,癌前病变及细胞增殖情况有关,提示有可能抑制细胞凋亡,促进肿瘤发生,或通过与其他基因的协同作用细胞增殖,导致癌变。

Abstract:Objective To study the expression and the roles of Bel?2 protein in the carcinogenesis of gastric carcinoma. Methods The expression of protein encoded by Bel?2 gene in 50 cases of gastric cancer and 10 normal gastric tissue by immunohistochemical staining was detected and compared according to Buke"s stages of tumors and precancerous lesions. Results The positive rates were 68.0%, 64.0%, 59.0% and 57.9% in advanced gastric cancers, gastric cancer in early stages, gastric cancers in early stage with intestinal metaplasia and dysplasia respectively, all being higher than that (20.0%) in normal gastric tissue. According to the degree of differentiation, high differentiatd gastric cancer had a positive rate of 82.4% > moderate differentiated gastric cancer(50.0%) > low differentiated gastric cancer (17.6%). The extent of proliferative zone could be seen in the tissue of surrounding cancer tissue. The expression of Bel?2 protein was strongly positive in proliferative cells and glands. Conclusion Bel?2 protein is an important pro oncogene and play an important role in the carcinogenesis.The expression of Bel?2 protein in gastric carcinoma is associated with the degree of differentiation, precancerous lesion and proliferative cells. It suggests that Bel?2 may lead to carcinogenesis either by inhibiting apoptosis or by inducing cell proliferation together with other genes.
