豚鼠心房肌细胞的分离及其离子流记录

摘    要:为探讨豚鼠心房肌细胞的分离方法及其离子流记录,采用酶解技术分离豚鼠心房肌细胞,应用膜片钳全细胞记录技术,记录细胞膜L型钙流(L-I_(Ca))及延迟整流性钾流(I_K)。结果获得了具正常电生理活性的单个细胞及高阻封接的形成,成功记录了L-型钙流和延迟整流性钾流。认为本方法分离的单个心房肌细胞具正常的电生理活性,可用于研究心房肌细胞离子通道活性。

Abstract:In order to study methed for isolating single atrial myocyte and its ionic current recondihg, single guinea-pig atrial myocyte obtained by enzyme isolated method and patch-clamp technique was adopted to record L-Ica and IK. As a result single guinea-pig atrial myocyte with normal electrophysiologic activity was obtained and its L-Ica and IK were successfully recorded with patch-clamp technique. Authors have come to conclussion that enzyme isolated method is a success in obtaining single guinea-pig atrial myocyte with normal electrophysiologic activity, and study of ionic channel activity with patch-clamp technique will become possible.
