单侧肾脏结扎术+链脲佐菌素诱导糖尿病肾病动物模型建立方法的探讨

摘    要:目的:探讨采用单侧肾脏结扎术 腹腔注射链脲佐菌素(STZ)诱导的方法建立糖尿病肾病(DN)模型的可行性。方法:采用单侧肾脏结扎术 腹腔注射STZ(50mg/kg)及单侧肾脏结扎术 尾静脉注射STZ(40mg/kg)的方法建立糖尿病肾病模型,比较2模型组成模率及大鼠血糖的稳定性。在此基础上以肌酐清除率(Ccr)为指标,探讨腹腔模型组大鼠是否存在有健侧肾脏肥大,与正常对照组作比较。结果:2模型组模型成功率无显著差异(P>0.05),但CF组10d平均血糖变异系数小于CW组。CF组大鼠尿蛋白定性呈阳性,Ccr显著升高,与正常对照组比较(P<0.05)。结论:单侧肾脏结扎法 50mg/kg体重腹腔注射STZ大鼠糖尿病肾病模型可以用来作为研究糖尿病肾病的理想工具。

Abstract:Objective:This study was conducted to explore unilateral nephritic ligation and streptomycin(STZ)abdomen injection inducing diabetic nephropathy(DN)of rats.Methods:Using unilateral nephritic ligation and STZ abdomen injection(50 mg/kg)and unilateral nephritic ligation and STZ venoclysis of the tail(40 mg/kg)induced STZ DN of rats.Comparing the rate of the developed model and the stability of blood sugar of two groups of DN rats,and by detecting creatinine clearance(Ccr)of the rats to find the development of nephrohypertrophy of the other side of the rats by unilateral nephritic ligation and STZ abdomen injection.Results:Compared of two groups of DN rats,the rate of the developed model in two DN rats is undifferentiated(P>0.05).But compared with rats of STZ venoclysis of the tail,CV of blood sugar was lower in rats of STZ abdomen injection.Compared with normal rats,Ccr was higher in rats of STZ abdomen injection.(P<0.05).Conclusion:Unilateral nephritic ligation and STZ abdomen injection(50 mg/kg)inducing DN of rats are a good way to study DN.
