等位基因特异性PCR技术在DEL表型基因分型中的应用

目的：建立DEL表型的基因分型方法，用于快速鉴定DEL表型。方法：用RH Box基因分型方法鉴定标本的基因型，用血清学方法确定标本的DEL表型。根据RHD1227A等位基因序列设计等位基因特异性．聚合酶链反应（allele specific-polymerase chain reaction，AS—PCR），用于DEL表型基因分型，结果同血清学的方法作比较，探讨临床应用的可行性。结果：28例RH Box基因分型为RHD^-／RHD^-的标本，血清学方法、基因分型方法DEL表型鉴定结果均为阴性；在25例RHBox基因分型为RHD^＋／RHD^-或RHD^＋／RHD^＋的标本中，用血清学方法检出的11例DEL表型标本，基因分型方法均扩增到867bp的1227A等位基因条带；1例血清学方法阴性，而基因分型方法检测为阳性，后经测序分析证实RHD1227A为阳性，分析此例标本可能为血清学方法漏检。这说明在鉴定DEL表型的方法中基因分型方法比血清学有更高的灵敏度。结论：AS—PCR方法可用于临床筛选Rh阴性人群中的DEL表型。

Applications of allele specific-PCR in genotyping method for DEL phenotype
