抑癌基因PTEN mRNA在白血病细胞中的表达及意义

摘    要:目的了解白血病细胞中PTEN基因的表达情况及其与临床的关系。方法采用RT-PCR法检测5个白血病细胞系,31例慢性粒细胞白血病(CML),87例初诊急性白血病(AL)包括59例急性髓系白血病(AML),26例急性淋巴细胞白血病(ALL),2例急性混合细胞白血病],21例完全缓解期急性白血病(AL-CR)患者骨髓标本中PTEN mRNA表达情况,并分析了与临床指标的相关性。结果PTEN基因在K562细胞中有表达,而在Kasum i-1、HL-60、U937、Nalm-1细胞中均无表达。CML组阳性率(61.29%)与正常对照组(78.57%)相比,差异无统计学意义(P>0.05),AL组和AL-CR组阳性率(分别为18.29%和42.86%)均低于正常水平(P<0.01和P<0.05),AL组阳性率又低于AL-CR组(P<0.05)。PTEN基因的阴性表达与外周血高白细胞计数(≥20×109/L)、染色体异常呈显著的正相关。结论AL患者存在PTEN基因的表达缺失,PTEN基因在AL的发病中可能起一定作用。

Abstract:OBJECTIVE: To explore PTEN gene expression and its clinical significance in acute leukemia. METHODS: The expression levels of PTEN mRNA in 5 leukemia cell lines, 87 patients with acute leukemias (AL), including 59 acute myeloid leukemia (AML), 26 acute lymphoblastic leukemia (ALL), and 2 acute hybrid leukemia, 21 AL in complete remission (AL-CR), 31 chronic myelogenous leukemia (CML) and 14 normal controls were assayed by RT-PCR. RESULTS: PTEN mRNA was detected in K562 cell line, but not in Kasumi-1, HL-60, U937, Nalm-6 cell lines. The expression ratio of PTEN mRNA between CML (61.29%) and normal control (78.57%) had no statistical difference (P > 0.05). The expression ratios of PTEN mRNA in AL (18.39%) and AL-CR (42.86%) were significantly lower than that in normal control (P < 0.01 and P < 0.05, respectively), AL also has a lower expression ratio than that of AL-CR (P < 0.05). The decreased level of PTEN mRNA had a positive correlation with poor-prognostic factors (high white blood cell count of > or = 20 x 10(9)/L and chromosome abnormality). CONCLUSION: There is down-regulated expression of PTEN gene in AL. PTEN gene may play a role in leukemogenesis.
