低氧状态下As_2O_3对人胃癌细胞SGC-7901抑制作用研究

摘    要:目的:探讨三氧化二砷(As2O3)在低氧状态下对体外培养的人胃癌细胞SGC-7901生长的抑制及凋亡的影响。方法:用氯化钴建立低氧模型,将细胞分为常氧组、低氧组、低氧加药(0.5、1.0、2.0、5.0、10.0μmol·L-1 As2O3)组,作用于胃癌细胞24、48、72h后用MTT法检测细胞光密度并计算抑制率,染色法检测细胞凋亡率。结果:作用24、48、72h后,低氧加药组中10.0μmol·L-1浓度抑制率分别为31.99%、55.98%和72.40%,凋亡率分别为(42.88±1.72)%、(56.48±1.48)%和(65.52±1.00)%,而低氧组则分别为4.04%、6.65%和8.11%,(2.08±0.28)%、(2.44±0.51)%和(2.76±0.67)%,两组比较差异具有显著性(P<0.05)。结论:低氧状态下As2O3对人胃癌细胞SGC-7901生长具有抑制和诱导凋亡作用,该作用可能是As2O3产生抗肿瘤效果的生物学基础。

Abstract:OBJECTIVE: To investigate the effect of arsenic trioxide (As2O3) on the growth inhibition and apoptosis of gastric cancer cell SGC-7901 cultured in vitro in hypoxia condition. METHODS: The environment of hypoxia was established by using cobalt chloride. The gastric cancer cells were divided into three groups: normal group, hypoxia group and hypoxia combined with arsenic trioxide (0.5, 1.0, 2.0, 5.0 and 10.0 umol. L^-1) (HAT) group. The optical density of gastric cancer cell was determined by MTT method and inhibition rate was calculated at 24, 48 and 72 h. The apoptosis rate of gastric caner cell was detected by using staining technique. RESULTS: After treated by 10.0 umol. L^-1 arsenic trioxide for 24, 48 and 72 h, the inhibition rates of SGC-7901 were 31.99%, 55.98% and 72.40% for HAT group and 4.04%, 6.65% and 8.11% for hypoxia groups, respectively; apoptosis rate were (42.88±1.72)%, (56.48±1.48)% and (65.52±1.00)% for HAT group and (2.08±0.28)%, (2.44±0.51)% and (2.76±0.67)% for hypoxia group, respectively. The difference between HAT group and hypoxia group was statistically significant (P〈0.05). CONCLUSION: Arsenic trioxide can inhibit growth of human gastric cancer cell SGC-7901 and induce its apoptosis in hypoxia condition. The mechanism may be the biological basic of arsenic trioxide to play a role of anti-tumor.
