摘    要:目的:探讨利用PCR—SSCP快速检测和鉴别临床病原菌的方法。方法:采用细菌保守的16s rRNA基因为模板,以PCR—SSCP方法检测病原菌,并以普通PCR法和PCR—RFLP法作比较。结果:普通PCR法很难从电泳图谱中鉴别细菌;PCR—RFCP法电泳图谱虽呈多态性,但仍有部分细菌图谱相同和相似,不易区分;经PCR—SSCP电泳出来的细菌图谱差异明显,可以互相区分。结论:PCR—SSCP技术能快速简便地检测、鉴别病原菌。

Abstract:Objective:To find the method of the identifing and detecting clinical pathogens for clinical diagnosis and treatment.Methods:A polymerease chain reaction targeted conserved sequence of enbacterial 16s rRNA gene,combined with single strand configuration polymorphism(SSCP) was used to detect clinical pathogens,and compared with common PCR and PCR RFLP(restrict fragment length polymorphism).Result:common PCR was very difficult to identify pathogens from electrophoresis map,PCR RFLP expressed polymorphsm,but there were either the same or similar map among some pathogens.However,there were significantly different bands in all the pathogens by PCR SSCP.Conclusion:PCR SSCP analysis can rapidly and conveniently identify bacterial pathogens.
