不同致呕吐物质对大鼠延髓组织5-HT表达的影响

目的：通过对大鼠给予不同致呕吐物质的刺激,研究大鼠在接受不同呕吐物质刺激后其延髓组织5-羟色胺（5-HT）的变化,为中医学＂呕吐＂证发生的病理病机＂胃气上逆＂的理论,提供分子生物学依据。方法：Wistar健康成年大鼠32只,随机分为4组,即对照1组,藜芦组,对照2组,顺铂组,每组8只。对照1组以生理盐水20mL/kg灌胃;藜芦组给予藜芦浓缩液灌胃,计量为20mL/kg,每毫升浓缩液相当与0.28g生药;对照2组腹腔注射生理盐水10mL/kg,顺铂组以浓度1mg/1mL顺铂注射液10mL/kg腹腔注射。给药6h后进行标本采集,取大鼠延髓。采用高效液相色谱法检测大鼠延髓组织中5-HT含量。采用免疫组化法检测5-HT表达的变化。结果：与对照1组相比藜芦组大鼠延髓组织5-HT含量增高（P〈0.05）,表达上调（P〈0.05）。结论：藜芦可使大鼠延髓组织5-HT含量增高,表达上调。＂胃气上逆＂与延髓组织5-HT含量增加有关。

Study on 5-HT Levels in Rats Medulla Oblongata which Were Disposed by Different Medicine

Objective：To study the changing of 5-HT levels in medulla oblongata when rats were disposed by different medicine that can make people vomit and provide experimental basis for detecting the pathological mechanism of reverse ascending of Stomach-Qi.Method：Thirty-two healthy Wistar rats were randomly divided into four groups：control 1 group,hellebore group,control 2 group,cisplatin group,8 rats per group.The control 1 group were given saline via intragastric administration（i.g.）,hellebore group were given concentrated solution of hellebore via（i.g.）.The control 2 group were given saline via intraperitonealinjection（i.p.）,cisplatin group were given cisplatin via（i.p.）.6 hours later,medulla oblongata was taken out of rats.5-HT were observed by high efficiency liquid chromatography,5-HT levels were measured with radioimmunoassay.Results：5-HT concentration in the bulbus medullae of hellebore group is more than those of control 2 group.Conclution：Hellebore increases 5-HT levels in the bulbus medullae;Reverse ascending of Stomach-Qi is associated with the increasing of 5-HT a levels in the bulbus medullae.
