人参皂苷Rg1抗同型半胱氨酸诱导人脐静脉内皮细胞凋亡的作用及相关机制研究

摘    要:目的探讨人参皂苷Rg1抗同型半胱氨酸(Hcy)诱导人脐静脉内皮细胞(HUVECs)凋亡的作用及相关机制。方法体外培养HUVECs细胞,采用琼脂糖凝胶电泳和末端脱氧核糖核苷酸转移酶介导的缺口末端标记法(TUNEL)染色检测细胞凋亡情况,逆转录-聚合酶链反应(RT-PCR)和Western blotting分析各组内皮型一氧化氮合酶(eNOS)mRNA和蛋白质的表达。结果琼脂糖凝胶电泳显示:与对照组和Rg1组比较,Hcy组可见特征性DNA的"梯状"条带。TUNEL染色显示:与对照组和Rg1组比较Hcy组细胞凋亡率增加差异有统计学意义(P<0.01);RT-PCR显示:与对照组和Rg1组相比,Hcy组eNOS mRNA水平表达降低差异有统计学意义(P<0.01);与Hcy组相比,Hcy+Rg1组eNOS mRNA水平表达增加差异有统计学意义(P<0.01),但仍低于对照组和Rg1组,差异有统计学意义(P<0.01)。Western blotting显示:与对照组和Rg1组相比,Hcy组eNOS蛋白水平表达降低差异有统计学意义(P<0.01);与Hcy组相比,Hcy+Rg1组eNOS蛋白水平表达增加差异有统计学意义(P<0.01),但仍低于对照组和Rg1组,差异有统计学意义(P<0.01)。结论人参皂苷Rg1能够抑制Hcy诱导的人脐静脉内皮细胞凋亡,此作用可能与上调血管内皮细胞eNOS水平有关。
